A role for the TGFβ-Par6 polarity pathway in breast cancer progression

Abstract: The role of polarity signaling in cancer metastasis is ill defined. Using two three-dimensional culture models of mammary epithelial cells and an orthotopic mouse model of breast cancer, we reveal that Par6 signaling, which is regulated directly by TGF␤, plays a role in breast cancer metastasis. Interference with Par6 signaling blocked TGF␤-dependent loss of polarity in acini-like structures formed by non-transformed mammary cells grown in three-dimensional structures and suppressed the protrusive morphology o… Show more

“…Migration of cells is associated with changes in cell architecture, suggesting that cell polarity proteins play important roles during this process. Recently, the polarity protein Par6 has been shown to play a critical role during TGFbinduced EMT 33,34 . In addition, loss of Hugl-1 also is associated with EMT in melanoma cells; restoration of expression Hugl-1 restores epithelial cell polarity while also inhibiting cell migration 35 .…”

“…PAR6 has been established as a causal factor for breast cancer epithelial-messenchymal transition (EMT) through transforming growth factor-beta (TGF-b) signaling. Mutated PAR6 attenuates TGF-b signaling in mice and suppresses lung metastasis of mammary tumors [12]. Loss of LGL1, a mammalian ortholog of fly L(2)GL, disrupts neural progenitor cell differentiation in the ventricular region of the mouse cerebellum, and promotes proliferation [13].…”

“…In a similar fashion, signaling induced by TGF-β1 destabilizes PAR6 and affects both E-cadherin expression and localization of the PAR complex. 102,103 Because Numb establishes a dynamic link between AJs and tight junctions, loss of Numb upon HGF stimulation could affect both structures simultaneously during EMT.…”
