“…An essential criteria for the development of the ELISA is the presence of two or more antibody‐binding sites on the target proteins. 14 Generally, monoclonal antibodies, when used as coating proteins, are known to provide lower background and higher specificity than polyclonal antibodies. 13 However, for the detection of a complex antigen such as BROS, the use of monoclonal antibodies does not provide the required sensitivity.…”

“…The rabbit antibody IgG-coupled glass rods thus prepared were tested by the sandwich procedure. As shown in Table 2, p-D-galactosidase activity bound to the glass rods was decreased by 3,11,38 and 53 % when 2 mg/ml solution of the rabbit antibody IgG fraction for coupling was diluted 2-, 6-, 10-and 20-fold, respectively, with non-specific rabbit IgG fraction (2 mgiml). When 200 pg/ml solution for coupling was diluted 2-, 6-, 10and 20-fold, it was also similarly decreased by 0, 10, 33 and 43 respectively.…”

“…Cellulose [3] and Sepharose 4B [4-71 have been used as the solid phase for the sandwich immunoassay. To wash fine particles of cellulose or Sepharose 4B during the assay procedure, the supernatant fluid after centrifugation has to be removed carefully.…”

“…They are able to bind to the Fc part of IgG, especially if the shape of IgG is modified by aggregation or by specific reaction with an antigen [3], RFs can alter EIA and immunofluorescent tests. Sev eral mechanisms have been described: specific binding with IgG conjugate itself [4], specific binding both with serie IgG bound to a fixed antigen and an anti-IgM con jugate [5] or also nonspecific binding with components of the kit [6].…”
