A Serum Metabolomic Signature for the Detection and Grading of Bladder Cancer

Abstract: Bladder cancer has a high incidence and is marked by high morbidity and mortality. Early diagnosis is still challenging. The objective of this study was to create a metabolomics-based profile of bladder cancer in order to provide a novel approach for disease screening and stratification. Moreover, the study characterized the metabolic changes associated with the disease. Serum metabolomic profiles were obtained from 149 bladder cancer patients and 81 healthy controls. Different ensemble machine learning models… Show more

“…The low levels of glutamine are consistent with the well-known "glutamine addiction" of cancer cells [34] reported in bladder cancer [35], lung cancer [36], and glioma [37]. This is due to the need for cancer cells to sustain the production of acetyl-coenzyme A to synthetize fatty acids when pyruvate is converted to lactate.…”

“…Metabolomic impairments of cancer cells represent a well-known cancer hallmark [ 44 ]. These impairments are widely regarded as a robust basis of cancer metabolic profiles [ 35 , 36 , 45 ].…”

“…Extraction, purification, and derivatization of serum samples were performed using the MetaboPrep GC kit (Theoreo srl, Montecorvino Pugliano, Italy) as reported in Troisi et al [ 28 , 29 , 32 , 33 , 34 , 35 , 36 ]. This sample preparation kit results in untargeted metabolomic profiles.…”

“…Statistically, in case of 80% of patients tumor do not spread outside of the bladder wall. BC can also be classified by histology as low-grade (LG) tumor that rarely spread from their primary site, and high-grade ones (HG) that are more aggressive and invasive 3 .…”
