A Simple Enzymic Digestion Procedure of Intact Tissue Samples in Pharmacokinetic Drug Analysis

Abstract: A simple procedure is described for the enzymic digestion of intact solid tissue samples. The samples are digested in an enzyme suspension containing collagenase and subtilisin Carlsberg without additional grinding or homogenizing. Bacterial growth during digestion is reduced by addition of an antibiotic solution. The resulting digests contain only very small tissue fragments. The total digestion is carried out at physiological conditions of temperature and pH, which makes the procedure suitable for inclusion … Show more

“…Initial screening tests for drugs other than volatiles were performed using enzyme‐linked immunoabsorbant assay (ELISA) tests (ISOTEL, Ithaca, NY). For solid tissues, enzymatic digestion was performed before ELISA (11). For volatile substances a gas chromatographic procedure was used (12).…”

“…Enzyme Tissue Digestion. Tissue extraction of sulfluramid and deethylsulfluramid was carried out using a procedure based on enzyme digestion of tissue as described by de Groot and Wubs (1987) with the exception that a higher collagenase concentration was used. Each tissue sample was minced with a scalpel blade and placed into a 50-mL centrifugation tube with a Teflon-lined screw cap.…”

“…Platinum was determined by atomic absorption spectroscopy (AAS) with electrothermal atomization in a graphite furnace using an Instrumentation Laboratory (Lexington, MA, USA) 451 Video AAS spectrometer,% DXR by a high performance liquid chromotographic procedure with fluorescence detection." The tissues were digested by means of an enzymatic digestion procedure 26. In addition, the concentrations of CDDP and DXR in kidney tissue were determined and used as reference values to make sure that the i.p.…”
