SUMMARYThe addition of 0.4 mmquinacrine to exponential cultures of Bacillus cereus led to the complete inhibition of growth, measured turbidimetrically. Certain metal ions such as Ca2+ and Mg2+ were partially effective in preventing growth inhibition, as was the polyamine spermine.Quinacrine inhibited most biosynthetic processes measured to the same extent as the reduction in the rate of growth. Thus the incorporation of amino acids into protein, that of diaminopimelic acid into bacterial wall, the accumulation of Kf and the uptake of nicotinic acid were diminished only in accordance with the decreased rate of turbidimetric increase. DNA biosynthesis was decreased to a lesser extent than was turbidity and continued even after growth had ceased altogether, thus indicating less inhibition of this process than that of protein formation. RNA synthesis, on the other hand, was selectively inhibited in these cells. The depressant effect was most pronounced when ribosomal or rRNA formation was examined. RNA isolated from drug-treated cells still exhibited messenger function. An additional effect of quinacrine on the uptake of certain nucleic acid bases is also postulated. The relative content of ATP in the cells was considerably increased by drug treatment.It appears that quinacrine did not exert its major action on DNA biosynthesis but selectively blocked RNA formation while that of DNA was permitted to continue.

2',3'-Dideoxyadenosine (DDA) was shown not only to possess antibacterial activity in vitro against a variety of Enterobacteriaceae, but also to be effective in vivo, DDA was active in experimental mouse infections by the oral route against 5 Salmonella strains, 2 of 3 Arizona strains, 5 of 7 Citrobacter strains, 3 of 8 Klebsiella strains, 3 of 5 Escherichia strains, 1 of 3 Shigella strains, and 3 of 15 Serratia strains at concentrations generally well below the toxic level. Closely related compounds, with the exception of 2',3'-dideoxyinosine, were found to be inactive in vivo, indicating that a high degree of structural specificity was required for activity. The synthesis of deoxyribonucleic acid was inhibited by DDA in those strains susceptible in vitro to DDA, whereas ribonucleic acid and protein syntheses were not affected. The concentration of DDA which inhibited bacterial deoxyribonucleic acid synthesis by 50% was calculated based on the relative rates of deoxyribonucleic acid synthesis in ;the absence and in the presence of DDA. This value correlated well with the minimal inhibitory concentration determined by the in vitro broth dilution assay but not always with in vivo activity determined by the mouse protection test.

Sodium arsenite (0–4 mm) inhibited the growth of exponentially growing Bacillus cereus, whereas sodium arsenate was growth‐inhibitory only at 10 mm concentration. When the phosphate concentration was reduced sharply, 3.3 mm sodium arsenate inhibited growth, whereas the inhibitory effect of arsenite was independent of phosphate concentration. Neither arsenite nor arsenate produced any specific effects on the incorporation of precursors into ribonucleic and deoxyribonucleic acids, protein or cell wall, in support of the concept that their actions were at stages of energy utilisation rather than biosynthesis of macromolecules. Although some 74arsenate may have been bound by cells during inhibition of growth, 74arsenite was selectively and potently concentrated in micro‐organisms. There was no evidence for the formation of arsenic‐containing nucleic acids nor were labelled organic intermediates found. No instance of interconversion between pentavalent and trivalent states of arsenic could be established, and it was concluded that both compounds inhibited the growth of B. cereus by separate mechanisms.
