Cartilage bone-marrow extract has stimulated the collagen formation of articular as well as sternal cartilage collagen in chick embryo. Collagen biosynthesis has been stimulated also in other investigated tissues, i.e. in cornea and sclera of chick embryo as well as in sponge granuloma of rats, where mainly formation of collagen type I was stimulated. Glycosaminoglycans formation has also been increased after administration of cartilage bone-marrow extract.

The effect of cartilage bone marrow extract (CBME) on the metabolism of collagen and proteoglycan in crude embryo and human cartilage was investigated. While in chick embryos’ articular cartilages the increased levels of radioactivity in collagen and proteoglycan were observed, in normal human cartilage only slight changes were detected. In osteoarthrotic cartilages, however, a marked increase in radioactivity was found in collagen and in the proteoglycan fraction containing the glycoprotein link. A blockade of collagenolysis by CBME treatment is suggested.

“…The excised corneae were shaken separately in 100 ml o f 0.1 m sodium sulphate at 4°C overnight and afterwards in 100 ml o f distilled water for several hours to remove free radioactive inorganic sulphate [Dohlman et al, 1968], The corneae were then dried, digested with papain (Wor thington Biochemical Corporation, Freehold, N.J.), and the enzymic digest submitted to chromatography on cetylpyridinium chloride cellulose [Praus, 1970]. The keratan sulphate and chondroitin sulphate fractions were analyzed for hexosamine by the Elson-Morgan reaction and assayed for radiactivity using an liquid scintillation counter.…”

Soft poly(hydroxyethylmethacrylate) gel contact lenses soaked with a solution containing either radioactive sulphate or D-glucose-¹⁴C were applied to the rabbit eyes for time intervals ranging from 10 min to 24 h. It was estimated that the radioactive substances were released very rapidly from the contact lens; about 90% within the first hour of application. Radioactivity was then detected in the cornea, aqueous humour, crystalline lens, and vitreous in amounts decreasing with the duration of the experiment. A stepwise accumulation of radioactivity in the eye tissues and fluids was observed with sulphate but not with glucose.
