A Simple Method for the Acquisition and Transmission of Brassica Yellows Virus from Transgenic Plants and Frozen Infected Leaves by Aphids

Abstract: Brassica yellows virus (BrYV) is a tentative species of the genus Polerovirus, which occurs widely, and mostly damages Brassicaceae plants in East Asia. Because BrYV cannot be transmitted mechanically, an insect-based transmission method is required for further virus research. Here, a reliable and unrestricted method is described, in which non-viruliferous aphids (Myzus persicae) acquired BrYV from transgenic Arabidopsis thaliana, harboring the full-length viral genome germinated from seeds and its frozen leav… Show more

“…To test whether TPT is involved in biotic stresses like viral infection. Col-0 and tpt3 (a T-DNA insertion mutant of tpt (allele tpt3 , SALK_093334) ( 49 )) were inoculated with BrYV (transmitted by aphids) ( 39 ). In comparison to WT (Col-0), the abundance of BrYV CP, MP protein and RNA were significantly increased in virus-infected tpt3 plants (Fig.…”

“…Total protein and RNA were extracted from treated areas on the leaves, and viral proteins including CP and MP and RNA as representatives of virus abundance were analyzed by western blotting and RT-qPCR respectively. The detailed method for aphid-mediated transmission of BrYV was described previously ( 39 ). For TuMV-GFP and CMV infection assays, four-week-old Arabidopsis seedlings were inoculated mechanically with sap from TuMV-GFP-infected N. benthamiana leaves (ground in phosphate buffer [pH7.4]) or purified CMV virions (diluted to 5 ng/μL) as previously described ( 94 ).…”

“…Aphids consume plant sap, which can result in stunted growth. They can also transmit various viruses, including those that affect the Brassicaceae family [ [23] , [24] , [25] , [26] , [27] , [28] ]. In addition, aphids excrete honeydew, which can lead to fungal growth [ 10 ].…”

“…When a 35S promoter-derived expression cassette containing the full-length cDNA of Br-C was successfully transformed into the Arabidopsis thaliana Col-0 ecotype, two lines, 111 and 412, of these transgenic Arabidopsis plants were generated for further study [13]. M. persicae has been reported as the vector of BrYV and a simple novel method was established for the acquisition and transmission of BrYV from transgenic line 412 Arabidopsis plants or frozen infected leaves [14]. Polerovirus particles are composed of the major coat protein (CP) and a few copies of a minor readthrough protein (RTP) which is a fusion protein with the CP at its N-terminus and the readthrough domain (RTD) at its C-terminus.…”
