A Single Aspiration of Rod-like Carbon Nanotubes Induces Asbestos-like Pulmonary Inflammation Mediated in Part by the IL-1 Receptor

Abstract: Carbon nanotubes (CNT) have been eagerly studied because of their multiple applications in product development and potential risks on health. We investigated the difference of two different CNT and asbestos in inducing proinflammatory reactions in C57BL/6 mice after single pharyngeal aspiration exposure. We used long tangled and long rod-like CNT, as well as crocidolite asbestos at a dose of 10 or 40 µg/mouse. The mice were sacrificed 4 and 16 h or 7, 14, and 28 days after the exposure. To find out the importa… Show more

“…62 MWCNTs are associated with MCM development in the exposed lungs. 34,35,43,63,64 In order to evaluate the impact of HA functionalization in MW-induced MCM development, we analyzed mucus-producing goblet cells in lung sections and quantified chemical mediators involved in the MCM phenotype. Scoring criteria were based on published methodology and are detailed in Supporting Information methods.…”

“…Surprisingly, in thymi of aged mice, spiculate crystal-containing macrophages were located in thymic medulla. The crystalline material is reminiscent of Ym1-like CharcotLeyden crystals, which are linked to higher IL-1β secretion and exuberant innate immune response (31,32). Importantly, the FGF21 coreceptor βKlotho is not expressed in macrophages (Fig.…”

“…Interestingly, electron microscopy analysis revealed that macrophages in the aging WT thymi contained spiculate crystalline material reminiscent of Charcot-Leyden crystals (Fig. 2 F and G) (31,32), phagocytosed lipid droplets, and large protein aggregates, suggesting defective autophagy. There were also enlarged lysosomes with electrodense material (Fig.…”

“…Nanomaterials used in this study were previously characterized and reported in NANOATLAS (Vippola et al, 2009) as well as in our previous publications (Kinaret et al, 2017a;Rydman et al, 2015;Rydman et al, 2014;Palomäki et al, 2015). Providers and material characteristics are reported in Table 1.…”

“…We and others already showed that ENMs are able to exert toxic effects on the respiratory system, by using animal exposure models (Kinaret et al, 2017a;Rydman et al, 2015;Rydman et al, 2014). Apart from the pathological responses, ENMs MOA has been investigated in different tissues (Kinaret et al, 2017b;Fröhlich, 2017;Costa and Fadeel, 2016), by analyzing the molecular perturbations that these materials are able to induce on the normal transcriptional program of exposed cells and tissues.…”
