RESUMOEste trabalho propõe uma aula prática para estudar a digestão de proteínas por proteases presentes em fármacos com digestivos enzimáticos, com simulação das condições de pH, temperatura e agitação do estômago e do intestino através do uso do dissolutor. Este experimento permite que os alunos dos primeiros semestres da graduação aprendam a utilizar equipamentos simples e técnicas fundamentais para o estudo da digestão de proteínas. Este experimento permite ainda a discussão de conceitos sobre estrutura e função de proteínas, pH e temperatura adequados e ainda conceitos iniciais de enzimologia. ABSTRACTThis study proposes a lab class to study protein digestion by proteases found in pharmaceutical preparations that contains digestive enzymes. The stomach and intestine pH, temperature and motion conditions were simulated using a dissolutor. This experiment allows students in the first terms of the undergraduate courses in Health Sciences, to learn how to use simple equipment and techniques that are fundamental to the study of protein digestion, and to learn the basic concepts of enzymology, since they will discuss concepts of the structure and function of proteins and suitable pH and temperature values for enzymatic reactions.Endereço para contato: UNIARARAS Av. This article describes a one-semester project-based laboratory course for biochemistry students centered on the protein cytochrome c. Students learn a variety of protein purification techniques, including ammonium sulfate precipitation, dialysis, ultrafiltration, gel filtration chromatography, ion-exchange chromatography, and electrophoresis, by isolating cytochrome c. The oxidation-reduction properties of this relatively stable and forgiving protein are then characterized by UV-visible spectroscopy and electrochemistry. Electrophoresis is utilized to assess purity and determine molecular weight. Students isolate cytochrome c from bovine heart and yeast and compare their properties.

A laboratory curriculum has been designed for an undergraduate biochemistry course that focuses on the investigation of the green fluorescent protein (GFP). The sequence of procedures extends from analysis of the DNA sequence through PCR amplification, recombinant plasmid DNA synthesis, bacterial transformation, expression, isolation, and characterization of the protein by SDS-PAGE. A survey of participants found that the majority of them were performing most of these procedures for the first time and that participants found the exercises enjoyable and considered them a significant aid to their understanding of biochemistry, cell biology, and molecular genetics.
