A Small RNA, UdsC, Interacts with the RpoHII mRNA and Affects the Motility and Stress Resistance of Rhodobacter sphaeroides

Abstract: sRNAs have an important role in the regulation of bacterial gene expression. The sRNA, UdsC, of Rhodobacter sphaeroides is derived from the 3′ UTR of the RSP_7527 mRNA, which encodes a hypothetical protein. Here, we showed the effect of UdsC on the resistance of Rhodobacter sphaeroides to hydrogen peroxide and on its motility. In vitro binding assays supported the direct interaction of UdsC with the 5′ UTR of the rpoHII mRNA. RpoHII is an alternative sigma factor with an important role in stress responses in R… Show more

“…E.g. sRNAs were shown to make important contributions to the regulated formation of the photosynthetic apparatus and to regulation of stress responses in R. sphaeroides ( Mank et al, 2012 ; Adnan et al, 2015 ; Billenkamp et al, 2015 ; Müller et al, 2016 ; Peng et al, 2016 ; Eisenhardt et al, 2018 , 2021 ; Reuscher and Klug, 2021 ; Spanka et al, 2022 ). Many sRNAs were detected in the R. capsulatus transcriptome ( Licht et al, 2020 ), which are still awaiting functional characterization.…”

“…For instance, the wild-type lac promoter from E. coli is reported to function poorly in R. sphaeroides (26). While no inducible promoters have been examined in N. aromaticivorans to our knowledge, promoters inducible by light, oxygen, crystal violet, and IPTG have been tested in R. sphaeroides (26)(27)(28)(29). Although these foreign promoters can respond to an array of possible inducers, some inducers inhibit R. sphaeroides growth (crystal violet) (26) or substantially alter physiological processes of interest (light, oxygen).…”
