A Strategy for Designing a Peptide Probe for Detection of β‐Amyloid Oligomers

Abstract: Aggregation of β-amyloid (Aβ) is implicated in the pathology of Alzheimer's disease. Development of a robust strategy to detect Aβ oligomeric intermediates, which have been identified as significant toxic agents, would be highly beneficial in the screening of drug candidates as well as enhancing our understanding of Aβ oligomerization. Rapid, specific and quantitative detection, currently unavailable, would be highly preferred for accurate and reliable probing of transient Aβ oligomers. Here, we report the dev… Show more

“…The TEM image of Aβ monomers incubated for 5 days also clearly show the existence of the Aβ aggregates consisting of protofibrils and short fibrils (Figure 4; Panel Aβ only). Recently, dot-blotting with Aβ-specific antibodies was widely used to detect the spectrum of Aβ aggregates with different conformations [16], [27], [37], [57], [58], [59], [60]. OC is a polyclonal antibody that reacts with neurotoxic fibrillar oligomers, protofibrils and fibrils [16], [58].…”

“…These assays were performed to obtain an integrated picture of Aβ aggregation modulation by ER. With Aβ-specific antibodies, dot blotting has become an effective method for monitoring Aβ aggregate formation [1], [26], [27], [28], [29], [30]. Four Aβ-specific antibodies were utilized for this purpose.…”

“…8 Despite extensive research for many decades, it was only until recently that a brain imaging agent, Florbetapir, was approved by the Food and Drug Administration (FDA) to evaluate AD. 9 In recent years, however, there has been a paradigm shift with numerous reported efforts involved in the development of effective methods for Aβ oligomers detection, including oligomer-specific antibody, 10 oligomer-specific peptide-FlAsh system, 11,12 peptide-based fluorescent protein, 13 as well as the ELISA method. 14 Yet, these detection methods often involve laborious construction methods, complicated instrumentation, or a long testing time, which make them inconvenient to use.…”
