A Strong Protective Action of Guttiferone-A, a Naturally Occurring Prenylated Benzophenone, Against Iron-Induced Neuronal Cell Damage

Abstract: Abstract. Guttiferone-A (GA) is a natural occurring polyisoprenylated benzophenone with several reported pharmacological actions. We have assessed the protective action of GA on ironinduced neuronal cell damage by employing the PC12 cell line and primary culture of rat cortical neurons (PCRCN). A strong protection by GA, assessed by the 2,3-bis(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carbox-anilide (XTT)

“…In addition, oxidative stress is often implicated in neurodegeneration associated with various insults to neurons such as hypoxia and hypoglycemia [ 46 ] as well as with the neurodegeneration observed in Alzheimer's [ 47 ] , Parkinson's [ 48 ] and amyotrophic lateral sclerosis diseases [ 49 ] . Therefore, in association with our previous results [ 15 ] , the present study suggests that the plant-derived antioxidant GA has promising potential as a therapeutic agent against neurodegenerative diseases involving ROS and oxidative damage.…”

“…The chemical structure of GA consists of an electronically delocalized planar 6-membered cyclic system and 2 phenolic hydroxyl groups, which facilitate its free radical scavenging activity. In fact, the fi rst anodic voltammetric peak potential for GA of approximately 0.25 V [ 15 ] was almost 2 times lower than those observed for other well-known polyphenolic antioxidant compounds such as mangiferin (0.62 V), kaempferol (0.4 V) and morin (0.4 V) [ 36 , 40 ] . Considering the established relationships between low oxidation potential and high antioxidant power, these data support the antioxidant strength of GA [ 41 ] .…”

“…Reactions were carried out for 1 h at 37 °C in a shaking bath to ensure continuous flow of O 2 in the tubes and terminated by the addition of 1 ml of 2.8 % (w/v) trichloacetic acid followed by 1 ml 1 % thiobarbituric acid solution in 50 mM NaOH. After the heating at 80 °C for 20 min, absorbance at 532 nm was determined [10].…”

“…The neuroprotective action of KM-34 (0.001-10 µM) was tested on PC12 cells co-incubated for 24 h with FeSO 4 (5 mM) and AA (2 mM) [10]. After 24 h, the cell supernatant was removed and cell viability was measured.…”
