A Structural Model for Vinculin Insertion into PIP2-Containing Membranes and the Effect of Insertion on Vinculin Activation and Localization

Abstract: Summary Vinculin, a scaffolding protein that localizes to focal adhesions (FA) and adherens junctions, links the actin cytoskeleton to the adhesive super-structure. While vinculin binds to a number of cytoskeletal proteins, it can also associate with phosphatidylinositol 4,5-bisphosphate (PIP2) to drive membrane association. To generate a structural model for PIP2-dependent interaction of vinculin with the lipid bilayer, we conducted lipid-association, nuclear magnetic resonance, and computational modeling exp… Show more

“…A dramatic reduction in binding to either PIP2 or PIP3 was observed for CDGIΔC1 compared with CDGI (Figure C), indicating a role of the C1 domain in PIP2/PIP3 recognition. Next, we compared the PIP2‐binding specificity between the isolated C1 domains of CDGI and CDGII, using the Vinculin tail (Vt) domain, a protein known to preferentially interact with PIP2 as a positive control. Although very limited co‐sedimentation with PIP2 was observed for the typical, DAG‐binding C1 domain of CDGII, co‐sedimentation of Vt and the C1 domain of CDGI demonstrated association with PIP2 (Figure D).…”

“…Membrane bound talin recruits and activates vinculin. Vinculin localizes at the adhesion complex and interacts with PI(4,5)P2 to associate with the membrane 56 . Simulation data shows that PI(4,5)P2 is not required for vinculin localization at FAs but is needed for the activation of FA turnover during mechanotransduction processes 56 .…”

“…IP3 is a key regulator of the cytoplasmic Ca 2+ concentration by controlling a Ca 2+ channel IP3 receptor on the endoplasmic reticulum (ER) . In addition to being the precursor of DAG and IP3, PI(4,5)P2 is also a key messenger that regulates the polymerization of the actin cytoskeleton, plasma membrane tension, exocytosis, membrane fusion, activity of ion channels, and cell adhesion, etc . Thus, PI(4,5)P2 is of great importance during receptor signaling and cell activation.…”
