Polar lobes of eggs of the mud snail Ilyanassa obsoleta, detached at the "trefoil" stage of the first cleavage, are capable of incorporating labeled amino acid into protein. The rate of incorporation per unit volume is about half that of the whole egg. The ability to incorporate at a similar rate persists for at least 24 hours after isolation. The sum of the incorporation of isolated lobes and lobeless fragments approximates that of the whole egg. The results extend to this material (the anucleate polar lobe) evidence for long-lived messenger RNA. They suggest also that the demonstrated morphogenetic influence of the lobe, which is exerted primarily during cleavage, may be correlated with its ability to synthesize protein.

“…Table II, Part A shows that 77.1% of the total 35 S-methionine was partitioned into the lobeless egg and 16.9% into the polar lobe; similarly, Table II, Part B shows that these parts of the egg account for 76.8% and 12.9%, respectively, of the 35 S-methionine incorporated into protein. These distributions are not significantly different (F(l, 4)-1.58, P > 0.05) from the 84.2% of the volume of non-yolk cytoplasm in the lobeless egg and 16.5% in the polar lobe (Collier, 1957). From this distribution of radioactive amino acids, I infer that the egg's amino acid pool, a quantity not directly measurable in a small number of eggs or isolates, is distributed into the lobeless egg and the polar lobe in proportion to their cytoplasmic volume.…”

The relative rates of protein synthesis in the lobeless egg and polar lobe of Ilyanassa obsoleta were measured and found to be the same. Two-dimensional electrophoresis of 35 S-methionine in vivo labeled polypeptides extracted from the Ilyanassa egg and its isolates resolved the same set of polypeptides from the egg, lobeless egg, and isolated polar lobe. For those peptides detected it was concluded that (1) the lobeless egg and the polar lobe contain the same set of mRNAs, (2) these mRNAs are not differentially translated in either of these isolates, (3) the polar lobe does not regulate the synthesis of these proteins, either quantitatively or qualitatively, during early cleavage, and (4) the peptides synthesized in the isolated polar lobe were translated from mRNAs produced during oogenesis.It was shown by one-dimensional electrophoresis of 35 S-methionine-labeled basic peptides that the egg, lobeless egg, and isolated polar lobe synthesized, at approximately the same rate, two variants of HI histone, histones H2A, H2B, H3, and H4, and high mobility group peptides 14 and 17. Because these peptides were synthesized in the isolated polar lobe it was concluded that they were translated from maternal mRNAs.

The spectrum of low molecular weight compounds, in particular of ribonucleotides, within first cleavage stage embryos of the polar lobe-forming mollusc Nassarius reticulatus and the distribution of the compounds within the embryo at the trefoil stage of fust cleavage are analysed by means of capillary isotachophoresis after 0.5 M PCA extraction. The compounds which are found in the whole trefoil embryo (T), the lobeless part (LL), and the polar lobe (PL) respectively, and the mean quantities (nmol. lo-'; n=6) are: UTP (11.5, 4.8, 5.6), ITP (8.5, 3.6, 5.0) GTP (10.3, 3.0, 9.0), ATP (29.8, 13.4, 18.8) UDP (11.8, 3.4, 8.7). CTP(8.0, 3.1,4.5), GDP(5.3, 2.6, 3.4) ADP(16.5,6.1, 11.6), CDP(4.0, 1.4, 2.6), GMP (4.7,2.7,4.3), glucose+phosphate (G6P) (53.5,38.8, 13.0). These compounds appear to be localized in the non-yolk cytoplasmic pool. As the volume ratio of PL/LL for total volume and for non-yolk cytoplasmic volume is about 0.74 and 0.60 respectively, the concentration of all nucleotides in PL as compared to LL is significantly higher (HO, p
