A Sucrose-Enriched Diet Promotes Tumorigenesis in Mammary Gland in Part through the 12-Lipoxygenase Pathway

Abstract: Epidemiological studies have shown that dietary sugar intake has a significant impact on the development of breast cancer. One proposed mechanism for how sugar impacts cancer development involves inflammation. In the current study, we investigated the impact of dietary sugar on mammary gland tumor development in multiple mouse models, along with mechanisms that may be involved. We found that sucrose intake in mice comparable to levels of Western diets led to increased tumor growth and metastasis, when compared… Show more

“…Panc-02 cells injected orthotopically into the pancreases of syngeneic C57BL/6 (B6) mice can form hepatic metastases (25), and 4T1 breast cancer cells injected orthotopically into the mammary fat pads of syngeneic BALB/c female mice can form lung metastases (26,27). Panc-02 cells stably transfected with PPARD-shRNA-A (PPARD-shRNA-A-clone1 or -clone2) had significantly lower PPARD RNA and protein expression than did WT Panc-02 cells and Panc-02 cells transfected with control-shRNA (Supplemental Figure 3, A removed when it reached a predetermined size of 10 mm in the greatest diameter (Figure 4, E and G).…”

“…The challenges of the catabolism of sugars other than glucose may be widespread. For example, in humans, bypassing glucose-responsive regulatory mechanisms with fructose has been associated with diabetes (LĂȘ et al, 2009; Kolderup and Svihus, 2015) and cancer (Port et al, 2012; Jiang et al, 2016). …”

“…30,31 Clinical research is hindered by the variability in diets and the associated metabolic interactions that may occur when trying to study the effects of specific macronutrients. Weight and physical activity can also confound results of dietary studies.…”
