“…Considering the multi-year delay between collection of the blood sample at baseline and the imaging and cognitive assessments performed at follow-up 2, it is unclear how well this single measure of serum CRP represents individual's basal levels; this limitation may help explain the lack of associations found in our primary analyses. While our preliminary analyses showed stronger longitudinal associations between serum CRP concentrations taken multiple years apart in the UKBB data than reported in a recent meta-analysis ( Walsh et al, 2023 ), aggregating serum CRP measures from multiple time points could be of great value. Unfortunately, using multiple serum CRP measures was not possible in these analyses as CRP data at follow-up 1 were limited in number and follow-up 2 CRP data were unavailable.…”

“…Repeated measurement may be particularly important for inflammatory markers such as IL-6, which are subject to more diurnal variability than others (e.g., CRP; Pearson et al, 2003). Even though recent evidence supports the temporal stability of single measures of IL-6 at least over periods of days to months (Walsh et al, 2023) and there was more between-person than within-person variability in IL-6 in the current sample, repeated measures provided a more reliable assessment of IL-6. A further strength of the current study was the collection of blood samples for the assessment of IL-6 in participants' natural environments at a consistent time of day (the apex of diurnal variability), improving the ecological validity of the assessment.…”

“…Discordance between levels of systemic cytokines and their vascular expression may be partially explained by the complexity of the inflammatory process. Cytokines are local mediators, and their production can be compartmentalized ( 49 ). Patterns of cytokine production differ under various clinical conditions, particularly in low-grade chronic inflammation, and cellular mRNA may not correspond to circulation proteins.…”
