A Systems Biology Approach Identifies FUT8 as a Driver of Melanoma Metastasis

Abstract: SUMMARY Association of aberrant glycosylation with melanoma progression is based mainly on analyses of cell lines. Here we present a systems-based study of glycomic changes and corresponding enzymes associated with melanoma metastasis in patient samples. Upregulation of core fucosylation (FUT8) and downregulation of α-1,2 fucosylation (FUT1, FUT2) were identified as features of metastatic melanoma. Using both in vitro and in vivo studies, we demonstrate FUT8 is a driver of melanoma metastasis which, when silen… Show more

“…We next performed motif analysis on the top 150 SEs (Figure 5B) and identified JUN and FOSL1/2 (of the AP-1 TF complex), TEAD1/4, and TGIF1/2 (Figure 5H), all of which have been implicated in melanoma or other cancers (Agrawal et al, 2017; Ozanne et al, 2007; Verfaillie et al, 2015; Xiang et al, 2015). ChIP-seq analysis showed FOSL2 and TEAD4 enrichment at melanoma SEs and TEs (Figures 5I and S6C).…”

“…Several glycans on the tumor surface, such as Tn, sialyl-Tn, and T antigen, have been identified as being mediators of critical pathophysiologic events during the various steps of tumor progression, such as cell-cycle dysregulation, cell adhesion, invasion activity, and angiogenesis (9,10). For example, upregulation of fucosylation by fucosyltransferase 8 in melanoma drives invasion and metastasis (11). Tumor-associated carbohydrates have been linked to drug resistance in multiple in vitro models (12) and also indicated as prognostic markers by which to predict therapeutic efficacy in patients with breast cancer who have been treated with anthracycline-containing adjuvant chemotherapy (13).…”

“…Similar mechanisms have also been shown to regulate transcription of glycogenes producing tumor‐related glycans likely supporting malignant transformation . In addition to epigenetic‐based mechanisms, classical signaling‐mediated transcription factor activation has also been shown to regulate the transcription of glycogenes . For example, endoplasmic reticulum (ER) stress‐induced activation of XBP1 promotes expression of mannosidase and N ‐acetylglucosamine (GlcNAc) transferases (MGAT1, MAN1A1, and MGAT2) involved in N‐glycan maturation, thus shifting the glycosylation profile of secreted cargoes from oligomannose toward mature, hybrid, and complex type N‐glycans .…”
