A Targeted and Adjuvanted Nanocarrier Lowers the Effective Dose of Liposomal Amphotericin B and Enhances Adaptive Immunity in Murine Cutaneous Leishmaniasis

Abstract: PDD reduced the effective dose and toxicity of LAmB and resulted in elicitation of strong parasite specific T-cell responses. A reduced effective therapeutic dose was achieved by selective LAmB delivery to APC, bypassing bystander cells, reducing toxicity and inducing antiparasite immunity.

“…These studies demonstrated that G5 PPI-mannose conjugate is a low toxic AmB formulation, providing prolonged release of carried drug. AmB was also the object of interest for Daftarian et al [79]. They prepared G5 PAMAM bound to a targeting peptide-Pan-DR-binding epitope.…”

“…In addition to siRNA delivery, the ideal nanoparticles should provide sustained release of antimicrobials to inhibit the proliferation of pathogens, while simultaneously accelerating cell migration. Importantly, nanoparticles such as dendrimers were shown to be an effective way of delivering Ambisome, an antifungal drug, for treating cutaneous Leishmaniasis in a mouse model that included reduced drug toxicity (Daftarian et al, 2013). Nitric oxide-loaded nanoparticles have shown promising antimicrobial activity in a murine wound model (Krausz et al, 2015).…”
