A two-SNP IL-6 promoter haplotype is associated with increased lung cancer risk

Abstract: Background Aberrant expression of interleukin-6 (IL-6) may play an important role in lung carcinogenesis. Whether IL-6 promoter haplotypes are associated with lung cancer risk and their functions have not yet been studied. We tested the hypothesis that single-nucleotide polymorphism (SNP) and/or haplotypes of IL-6 promoter are associated with risk of lung cancer. Methods Two functional IL-6 promoter SNPs (-6331T>C and -572C>G) were genotyped in the discovery group including 622 patients and 614 controls, and… Show more

“…Among these SNPs, rs1524107, and rs2066992 were identified that have association with EC risk. Rs1524107 was identified that has associations with several disease, including rheumatoid arthritis (Li et al, ), major depressive disorder (Chen, Wu, Zhao, Fan, & Fang, ), Alzheimer's disease (Chen et al, ), adult‐onset asthma (Lajunen, Jaakkola, & Jaakkola, ), lung cancer (Chen et al, ) et al, however, we did not found any reports association with EC risk in Chinese han population. For rs2066992, Juo et al, researched the rs2066992 and endometriosis risk, however, there is no significance.…”

“…Chen et al . showed that a two-single-nucleotide polymorphism (SNP) CC (-6331C and -572C) IL-6 promoter haplotype was significantly more common among cases than among controls in both groups, indicating this haplotype is associated with increased lung cancer risk [ 25 ]. Therefore, comprehensive haplotype-based or multiple SNP-based strategies may provide more precise information on the genetic contribution of TNF-α or IL-6 to cancer etiology in the future [ 21 ].…”

“…Construction of luciferase reporter gene plasmids was performed as previously described (Chen et al, 2013). A 751-bp sequence of IL-6 gene was amplified from the genomic DNA of healthy volunteers homozygous for the -572 C allele.…”
