A Two-step Strategy for High-Value-Added Utilization of Rapeseed Meal by Concurrent Improvement of Phenolic Extraction and Protein Conversion for Microbial Iturin A Production

Abstract: Rapeseed meal (RSM) is a major by-product of oil extraction from rapeseed, consists mainly of proteins and phenolic compounds. The use of RSM as protein feedstock for microbial fermentation is always hampered by phenolic compounds, which have antioxidant property with health-promoting benefits but inhibit bacterial growth. However, there is still not any good process that simultaneously improve extraction efficiency of phenolic compounds with conversion efficiency of protein residue into microbial production. … Show more

“…Compared with the antibacterial activity of surfactin, iturin A is mainly fungicidal, and the amino acid composition of iturin A is complex. Meanwhile, iturin A yield produced by Bacillus is relatively low, although several pieces of research on fermentation process optimization have been conducted to improve its yield (Jin et al, 2015;Wang et al, 2021). Previously, by replacing the promoter of the iturin A synthetase cluster and deleting the negative regulator gene abrB in B. amyloliquefaciens, iturin A yield was increased by 1.81-fold (Xu et al, 2020).…”

“…The low synthesis capability of iturin A limits its application promotion, although several approaches of genetic engineering breeding and fermentation process optimization have been developed to improve iturin A production Jin et al, 2015;Zhang et al, 2017;Shi et al, 2018;Dang et al, 2019;Xu et al, 2020;Wang et al, 2021). To improve iturin A synthesis, fatty acid biosynthetic pathways were systematically strengthened in Bacillus amyloliquefaciens HZ-12, a wide-type strain with the synthesis capability of iturin A (Xu et al, 2020), and the synthesis modules of malonyl-CoA, malonyl-ACP, free fatty acid, and fatty acid-CoA were strengthened for iturin A production (Figure 1).…”

“…These seed meals are known either for their elevated polyphenolic concentrations, such as GSM, SBSM, and RSM or for their content in polyphenolic compounds with a strong antimicrobial and anti-inflammatory effect. Thus, they are promising agro-industrial byproducts to manage the piglet's weaning crisis (Tabasco et al 2011;Bouarab-Chibane et al 2019;Grosu et al 2019;Wang et al 2021;Czech et al 2022;Taranu et al 2022;Wang et al 2022).…”

“…Phenolic compounds of L. barbarum remain unused after the extraction of oil by pressing or solution extraction. Therefore, the extraction, identification, and quantification of active components in meals are important for the comprehensive exploitation of meals and the improvement of value-added by-products (Malešević et al, 2014;Wang et al, 2021). The embedded phenolic compounds hold promising nutritional and health benefits which can be exploited through suitable extraction methods.…”
