A yeast protein that influences the chromatin structure of UASG and functions as a powerful auxiliary gene activator.

Abstract: GRF2, an abundant yeast protein of Mr -127,000, binds to the GAL upstream activating sequence (UASc) and creates a nucleosome-free region of -230 bp. Purified GRF2 binds to sequences found in many other UASs, in the 35S rRNA enhancer, at centromeres, and at telomeres. Although GRF2 stimulates transcription only slightly on its own, it combines with a neighboring weak activator to give as much as a 170-fold enhancement. This effect of GRF2 is strongly distance-dependent, declining by 85% when 22 bp is interpose… Show more

“…Differential regulation may well occur at the transcriptional level as suggested by the complete divergence of the nucleotide sequences upstream of TKLl and TKL2. The promotor region of TKL2 displays a putative TATA box, some 220 bp upstream of the translational start site, and an inverse GRF2 consensus binding site (Chasman et al, 1990) located 190 bp upstream of this TATA box. For TKL2, a putative TATA box is found some 130 bp upstream of the translational start site.…”

“…It is multifunctional, as it is involved in transcriptional termination (20,21), binds to telomeres and centromeres (22), and plays a role in transcriptional regulation of a plethora of functionally unrelated genes transcribed by either polymerase I or II (18,(22)(23)(24)(25)(26). However, the direct transcriptional activation potential of Reb1p is marginal compared with specific activator proteins, but combinations of Reb1p binding sites with cognate motifs for weak transcription activators or dA⅐dT elements cause considerable synergistic effects (22,(27)(28)(29). Interactions between Reb1p and the basal transcription machinery are discussed as well (1,22,26,30,31).…”

Reb1p is encoded by an essential gene (32); however, the reason for its indispensability has not yet been established.…”

“…No sequence with significant similarity was found. Binding sequences for Raplp [26], Abflp [27], and Reblp [28], all global transcriptional activators [29], were not found. Furthermore, physiological roles could not be distinguished by carbon source, as cells harbouring AP20 or AP12R did not show any significant specificity towards carbon sources other than glucose and acetate, such as ethanol or lactate (data…”
