A γ-chain complex forms a functional receptor on cloned human lymphocytes with natural killer-like activity

Abstract: We have recently derived from human fetal blood (25 wks) a series of cloned cell lines that were selected for their ability to kill the conventional natural killer (NK) target cell K562. It was found that a fraction of these clones express CD3 proteins but not the monomorphic Ti alpha beta determinant recognized by WT31 antibody. One interleukin-2-dependent CD3+ WT31- clone, termed F6C7, was used for immunization of mice to generate monoclonal antibodies directed at a potentially novel recognition receptor. It… Show more

“…In contrast, circulating TCR g/d þ cells [36,37] and a minor subset of TCR a /b þ , CD8 þ cells which contains lymphokineactivated killer (LAK) cell precursors [38] express constitutively CD122 in the absence of CD25. This feature allows both cell types to proliferate following in vitro treatment with high-dose rIL-2.…”

“…It is now established that T lymphocytes of this subset express a TCR-y/S (35)(36)(37)(38) and are capable of performing nonrestricted (39,40) as well as MHC-linked specific cytolytic functions (41,42).…”

“…Several lines of evidence suggest that T cells are involved in some early, non-specific resistance mechanisms [54][55][56], particularly the localization of different subsets to different epithelia that are in contact with the external environment [24,[57][58][59][60] where they appear to function as activated cytotoxic T lymphocytes [61,62]. There is also evidence from both mice and humans [63,64] that T cells originating from the recirculating pool are reactive to self 60-70-kD heat shock proteins.…”
