A23187-induced pulmonary gas trapping and inflammation in the guinea pig

Abstract: A brief A23187 aerosol exposure produced prolonged airway obstruction with granulocyte accumulation in conscious guinea pigs. Aminophylline, atropine, pyrilamine, salbutamol, SC-41930 (a leukotriene B4 antagonist) and WEB 2086 (a platelet activating factor antagonist) were administered intravenously (i.v.) to evaluate their ability to prevent these changes. Inhaled salbutamol was also assessed. Aminophylline, atropine, and salbutamol (i.v. and aerosol) inhibited A23187-induced gas trapping (p less than 0.01). … Show more

“…The divalent cationic ionophore A23187 is a nonimmunogenic substance that induces bronchoconstriction and airway changes characterized by mild epithelial injury, peribronchial accumulation of granulocytes, and edema of the airways [1]. Such findings have been observed in patients with bronchial asthma, and may contribute to their characteristic airway hyperresponsiveness (AHR) [2].…”

“…Whereas the degree of airway hyperresponsiveness to substance P was similar for sulfuric acid-and A23187-exposed guinea pigs, the pattern of lung inflammation differed. Airway epithelial damage and peribronchiolar inflammation may have con tributed to the hyperresponsiveness we observe( with A23187 [6,8]. However, the minimal sulfuri~ acid-related histological changes suggests a dis C131 sociation between airway inflammation and increased responsiveness.…”

“…Stengel et al have shown that inhalation of calcium ionophore A23I87 results In airway obstruction with infiltration of peribronchial inflammatory cells, mainly eosinophils. in guinea pigs [2][3][4][5][6], Although an actively sensitized animal model frequently requires an antihistaminic drug to prevent systemic anaphylaxis, the A23187 inhalation requires no antihistaminic dmg to prevent systemic anaphylaxis. So an A23I87 inhalation asthmatic mode!…”
