Aberrant JAK/STAT Signaling Suppresses TFF1 and TFF2 through Epigenetic Silencing of GATA6 in Gastric Cancer

Abstract: Aberrant Janus kinase (JAK)/signal transducer and activator of transcription (STAT) signaling is crucial to the development of gastric cancer. In this study, we examined the role of STAT3 in the expression and methylation of its targets in gastric cancer patients. Results from RNA sequencing identified an inverse correlation between the expression of STAT3 and GATA6 in 23 pairs of gastric cancer patient samples. We discovered that the expression of GATA6 is epigenetically silenced through promoter methylation … Show more

“…60 Abnormal JAK/STAT signaling may be involved in epigenetic inhibition of GATA6 expression by promoter hypermethylation, which may further cause downregulation of trefoil factors 1 and 2 (TFF1/2) in GC. 61 In summary, GATA6 has multiple roles in the development of GC, and its gene amplification can be a high-risk factor for the development of GC. Enhancing the expression of GATA6 can also inhibit tumor migration and invasion through certain signaling pathways.…”

“…In addition, TFF2 and SST have been reported as tumor suppressors, and low level of TFF2 and SST were found in GC patients (66)(67)(68). TFF2 is a short peptide that secreted by gastric mucous neck cells and is helpful for cellular protection of gastrointestinal tract (69).…”
