Abnormal activity of transcription factors gli in high-grade gliomas

Abstract: Malignant transformation is associated with loss of cell differentiation, anaplasia. Transcription factors gli, required for embryonic development, may be involved in this process. We studied the activity of transcription factors gli in high-grade gliomas and their role in maintenance of stem cell state and glioma cell survival. 20 glioma cell lines and a sample of a normal adult brain tissue were used in the present study. We found the expression of gli target genes, including GLI1 and FOXM1, in all tested gl… Show more

“…TNC is overexpressed and correlated with tumor malignancy observed in many cancer types and is associated with glioma outcomes [34]. Previously, we have shown the increased levels of stem cell genes, including SOX2, OCT4, and GLI1 in these glioma cell lines [35,36] that coincide with the presence of the CD44 and TNC in all the exosome samples from CCM.…”

“…Secondly, it is a major vault protein (MVP), which is associated with nuclear pores, regulates nuclear-cytoplasmic transport and determines resistance to certain drugs [ 40 , 41 ]. It should be noted that its gene is a target of the transcription factor GLI1 [ 42 ], which according to our previous data is abnormally active in these glioma lines [ 36 ].…”

“…We hypothesized that the inherent glycosylation of such proteins and hydrolytic product (cellobiose) release from BC into biological system may be factors in triggering an immune response [73]. For this reason, we tested the biocompatibility of the enzyme using primary human glioblastoma available in local cell collection [46]. Glioblastoma cell line was chosen due to its highest inherent sensitivity towards toxic effects of a wide compound panel checked previously (data not shown).…”

“…For the cell viability test, human glioblastoma primary cell line # 15 from the laboratory cell culture collection [46] was used. Cells were cultured in DMEM/F-12 (1:1) medium, containing L-glutamine and supplemented with 10% fetal bovine serum (BioWest, Nuaillé, France) without antibiotics in 5% CO 2 at 37 • C. Cells were seeded in 4 parallel 96-well plates, seeding density was set at 1 × 10 4 cells per well.…”
