Abnormal development of motor neurons in perfluorooctane sulphonate exposed zebrafish embryos

Abstract: Perfluorooctane sulfonate (PFOS) is an environmental organic pollutant, the potential neurotoxicity of which is causing great concern in fish. In the present study, we examined the effects of PFOS on motor neurons, and investigated the potential toxicological mechanisms oxidative stress in zebrafish embryos. Six-hour post-fertilization (hpf) zebrafish embryos were exposed to 1.0 mg/L PFOS, then we examined the expression of alpha-tubulin, proliferating cell nuclear antigen (PCNA), cyclin-dependent kinase 5 (CD… Show more

“…Moreover, perfluorooctane sulfonic acid perturbs the functioning of motor neurones by altering α-Tubulin and proliferating cell nuclear antigen 56. It also induces brain oxidative stress by dysregulating cyclin-dependant kinase 5 and peroxiredoxin, and causes an inflammatory glial response 57.…”

“…Adult behavioral and cognitive disturbances were reported A C C E P T E D M A N U S C R I P T ACCEPTED MANUSCRIPT 17 after exposure of rodent pups to PFAAs, usually at high doses [241][242][243]. Abnormal motor neuron development and locomotion were seen in zebrafish larvae exposed to PFAAs [256,257]. Very little information is found about other PFASs than PFOA and PFOS, and other species, in particular wildlife.…”

“…Western blotting assay was conducted using approximately 300 larvae from each replicate (n ¼ 3) as described previously [28]. In total, 50 mg of each protein sample was loaded into each lane of a 12% sodium dodecyl sulfate-polyacrylamide gel, separated by electrophoresis, and then transferred to a polyvinylidene difluoride membrane.…”
