“…T cells for proliferation and adhesion tests were purified as described previously [30, 31] and were > 95% pure. Proliferative responses to coimmobilized OKT3 antibody and ECM proteins, and adhesion to ECM proteins following short‐term stimulation with phytohaemagglutinin (PHA) were assessed as reported previously [30, 31]. The results were expressed, respectively, as the arithmetic means of [ 3 H]thymidine (cpm) incorporated by triplicate cell cultures and optical density units of crystal violet bound by adhering cells.…”

“…Greatly enhanced binding to immobilised fibronectin has previously been described in a patient with Wegener's granulomatosis (14) and we therefore studied pbl from a patient with this disease. Enhanced binding to FITC-FN was recorded ( Fig.…”

“…The enhanced binding of PMA-stimulated T-lymphoid cells to immobilised fibronectin has been found to depend on events following receptor occupancy rather than from enhanced affinity (7). This could suggest that enhanced initial binding to fibronectin is not important for primed lymphocytes, and therefore gives rise to the question of whether the enhanced binding to immobilised fibronectin noted in pbl in chronic inflammatory diseases (11,14,30,39) is solely due to post-receptor occupancy events. Flow cytometric measurement of stimulated lymphocyte adhesion to soluble fibronectin and of pbl binding to soluble fibronectin in chronic inflammatory disease will help to clarify this issue.…”

“…In addition, these immune cells have to penetrate the ECM-rich capsule surrounding tumor masses. Activated T cells adhere strongly to type IV collagen but weakly to type I collagen, indicating that specific ECM components have a role in cell infiltration (10). Marked lymphocyte infiltration occurs outside the collagen I/ II-rich capsule of hepatocellular carcinoma or surrounding compressed hepatocytes near the capsule (11).…”
