Abrogation of Bronchial Eosinophilic Inflammation and Airway Hyperreactivity in Signal Transducers and Activators of Transcription (STAT)6-deficient Mice

Abstract: Signal transducers and activators of transcription 6 (STAT6) is essential for interleukin 4–mediated responses, including class switching to IgE and induction of type 2 T helper cells. To investigate the role of STAT6 in allergic asthma in vivo, we developed a murine model of allergen-induced airway inflammation. Repeated exposure of actively immunized C57BL/6 mice to ovalbumin (OVA) aerosol increased the level of serum IgE, the number of eosinophils in bronchoalveolar lavage (BAL) fluid, and airway reactivity… Show more

“…Physiological studies demonstrate an inhibition of pulmonary eosinophil infiltration, mucus overproduction and AHR elicited by OVA challenge in mice harboring the STAT6 À/À null allele. 6,15 Expression profiling of the lung tissue of STAT-6 deficient following multiple dose mIL-13 lung instillation did not reveal any genes that met the dual fold change and statistical criteria. Specifically, none of the IL-13 modulated genes in wild-type Balb/C mice were statistically different (Po0.05) from vehicle in the absence of STAT6 function.…”

“…However, because the response was induced via a STAT6 pathway, an overall Th2-type environment may be initiated during the RSV response (24,26). STAT6 pathways are involved in allergen-induced airway hyperreactivity and mucus production (53,54). It is likely that multiple factors contribute to the pathophysiology during airway disease, including the genetic background and the cytokine profiles that are generated locally.…”

“…In mouse models of allergic lung inflammation (41)(42)(43), contact hypersensitivity (44), and allergic diarrhea (45), STAT6 deficiency was accompanied by a lack of eosinophil influx into the sites of allergic inflammation. This effect may be at least partially due to defective eotaxin-1 production in these animals.…”
