Absence of Tolerance to Hypotensive Effects of Clonidine in Spontaneously Hypertensive Rats

Abstract: ABSTRACT-Development of tolerance to the hypotensive effects of clonidine was investigated in spontaneously hypertensive rats (SHR). Clonidine (125 pg/kg/day) was administered subcutaneously for 5 weeks using an osmotic infusion pump. During the whole infusion period, significant hypotensive and bradycardic effects were observed. Plasma clonidine concentrations were maintained relatively constant at about 2 ng/ml during the infusion period. On termination of treatment with clonidine, both the blood pressure an… Show more

“…The reduction in BP induced by flesinoxan is due to selective stimulation of central 5-HT1A receptors leading to the reduction of sympathetic tone. [17][18][19] Because conflicting results have been reported on the chronic antihypertensive effects of clonidine in conscious SHR 20,21 and because no data are available on the chronic effects of flesinoxan, we investigated the effects of these drugs after both acute (24 h) and chronic (4 weeks) infusion. However, because the modifications of carotid artery diameter may be partly or totally independent of modifications in BP level, we used a low and high dose of each drug.…”
