Abstract 1500: A novel nutrient mixture exhibits antitumor activity in human fibrosarcoma cell line HT-1080

Abstract: Introduction: Fibrosarcoma, an aggressive and highly metastatic cancer of connective tissue, is generally associated with poor prognosis. Cancer mortality usually results from tumor invasion of local tissue and metastasis to vital organs. We investigated the effect of a specific nutrient mixture (PB) of quercetin, cruciferex, curcumin, green tea extract and resveratrol on human fibrosarcoma cell line HT-1080 for viability, MMP expression, invasion, apoptosis and morphology. “…Although both docetaxel and chlorophyllin could inhibit cell migration alone, their combined treatment had a substantially stronger effect. There was no available study in literature investigating the effects of combined use of chlorophyllin with anticancer drugs on cell migration, however, these results were in line with other studies reporting that certain antioxidants and/or phytochemicals administration, such as chlorophyllin, reduced the cell migration in aggressive cancer cell lines (Lirdprapamongkol, Sakurai et al 2005, Roomi, Bhanap et al 2018, Dehghani, Kooshafar et al 2019). In a study on HT-1080 brosarcoma cells, it was shown that chlorophyllin dose-dependently reduced the cell migration, and this effect was highest at 50 µM chlorophyllin administration (Roomi, Bhanap et al 2018).…”

“…The inhibitory effects of PB were also observed in fibrosarcoma, which is an aggressive and highly metastatic cancer of connective tissue, and in melanoma, which causes the majority of skin cancer-related deaths, secondary to metastasis. The in vitro studies used human fibrosarcoma cell line HT-1080 [ 96 ] and melanoma A2058 cells [ 97 ]. The study showed that PB inhibited cell proliferation of human fibrosarcoma cells by 60% at 25 μg/mL and 80% at 50 μg/mL, and inhibited both MMP-2 and -9 secretion in a dose-dependent manner, with total inhibition at 50 μg/mL [ 96 ].…”
