Acid heteropolysaccharides with potent antileishmanial effects

Abstract: The present study investigated in vitro the effects of sulphated heterorhamnan (Go3), iota-/nu-carrageenans (G3d and EHW-I) and arabinogalactan (ARAGAL) polysaccharides on macrophage activation and inhibition of intracellular amastigotes of Leishmania (L.) amazonensis. All the sulphated polysaccharides (Go3, G3d and EHW-I) promoted increased nitric oxide production varying from 71 to 110%. The leishmanicidal activity of all compounds was compared to the inhibition effect of Meglumine Antimoniate at 300μg/mL (∼… Show more

“…Among these are ones derived from arabinogalactan and isolated from Anadenanthera colubrina , which induces increased nitric oxide by macrophages. 79 Alkaloids derived from Aspidosperma ramiflorum have shown activity against promastigotes. 80 Also, alkaloids derived from marine sources exhibit excellent in vitro antileishmanial activity, inhibiting DNA synthesis.…”

“…The parasite-macrophage interplay begins when macrophages detect parasite molecules through membrane receptors, which then stimulate the production of proinflammatory cytokines, such as TNF-α, IFN-γ, IL-12, IL-1β and of hydrogen peroxide, triggering the Th-1 type cellular response, which is related to the efficiency in the elimination of the intracellular parasites (7). However, in the amastigote form, the parasite is able to inhibit hydrolytic enzymes and deplete reactive species of nitrogen and oxygen while remaining protected and is capable of multiplying in the phagolysosome (8).…”

“…The antileishmanial effects of the polysaccharides occurred by different mechanisms: for sulfated polysaccharides, this may be an expressive increase in NO production by macrophages, while for arabinogalactan, it may be attributed to stimulation of O 2 •− and TNF production. 113 Polysaccharide polymers have also been used to deliver vanadium complexes, which are reported to be highly active metal-based drugs against a variety of trypanosomatids, including Leishmania species. 114 The immunomodulatory effects of the polysaccharides arabinogalactan, galactomannan, and xyloglucan, as well as their oxovanadium (IV/V) complexes, were evaluated on peritoneal macrophages exhibiting significant leishmanicidal activity (~60%-70%).…”
