Acquisition of antibodies to merozoite surface protein 3 among residents of Korogwe, north eastern Tanzania

Abstract: BackgroundA polymorphic malaria parasite antigen, merozoite surface protein 3 (MSP3), is among the blood stage malaria vaccine candidates. It is believed to induce immunity through cytophilic antibodies that disrupt the process of erythrocytes invasion by merozoites. This study aimed at assessing natural acquisition of antibodies to MSP3 in individuals living in an area with different malaria transmission intensity in preparation for malaria vaccine trials.MethodsThe study was conducted in individuals aged 0-1… Show more

“…The age-related increase of antibody levels observed for the antigens in Shewa Robit is in agreement with other studies [22,31,32]. Although not significant, antibody levels to the antigens in the study were higher in smear-negative group for P. falciparum infection in Boditi sera.…”

“…comm.). However, previous studies conducted in the same and nearby areas had shown that highlands had low malaria transmission compared to lowlands [12,33,34]. The current situation indicates that there is a remarkable changing pattern of malaria and these areas might be moving from control to pre-elimination levels [35].…”

“…Recombinant antigens were dissolved in 0.01 M PBS to concentrations: 0.1 μg/ml for (AMA-1 3D7, AMA-1 FVO, EBA-175 and GLURP-R2), 0.2 μg/ml for (MSP-1 42 FVO, MSP-1 42 3D7, and MSP-1 42 FUP), 0.5 μg/ml for (GLURP-R0, MSP-1 19 and MSP-3 FVO). These antigen concentrations were found to be optimal in previous studies for AMA-1, EBA-175, MSP-1 42 [1], GLURP-R0 [16-19], GLURP-R2 [16,18], MSP-1 19 [20] and MSP-3 [21]. Fifty microlitres of antigen solution was added to Immulon-4 plates (Dynex Technologies,Chantilly, VA).…”
