Acquisition of CD40 Expression During Murine B‐Cell Differentiation

Abstract: Expression of CD40 on mouse cells was investigated comparing the binding to cells of a monoclonal antibody against CD40, to that of a soluble fusion protein consisting of the extracellular domains of the mouse CD40-ligand. The analysis of a series of established cell lines failed to demonstrate expression of CD40 on pro- or pre-B cells, and indicated that CD40 expression was restricted to cells that had undergone productive heavy- and light-chain gene rearrangements, and expressed surface Ig. In cells from nor… Show more

“…All of these receptors are expressed in developing B cells and pre- and immature mouse B cell lines27,28, and all activate NF-κB and induce transcription of genes encoding pro-survival proteins. We therefore examined the proportions of Igλ + cells in the immature B cell compartments of mice lacking Bcl10 and MyD88, which mediate NF-κB activation downstream of these receptors1,29, or CD40.…”

“…To determine whether CD2:Stat6VT B cells are activated or simply have increased expression of Stat6-inducible genes, we analyzed the expression of other cell surface markers that indicate activation or maturation of B lymphocytes and are not regulated by IL-4. FACS analysis of freshly isolated B220 ϩ cells from wildtype and CD2:Stat6VT mice showed that CD40 expression, which is up-regulated on activated or mature B lymphocytes (43)(44)(45), was increased on transgenic B cells compared with wild type (Fig. 5A).…”

“…The low basal expression of CD40, together with its induced expression in inflammation, suggest that CD40 ligation is particularly relevant in inflammatory conditions; its increased expression by inflammatory mediators provides a mechanism for amplification in inflammation. The induction of CD40 expression upon podocyte differentiation may suggest that, as for other cell types [Castigli et al, 1996;Grandien et al, 1996;Ma and Clark, 2009], CD40 expression is a feature of cell maturation. How CD40 signaling regulates ECM homeostasis has been for a large part studied in the vascular system, but its role in other tissue remains ill understood.…”
