“…at the pituitary gland, upon hypothalamus or other central nervous system structure( 7), or even by a direct action upon the adrenal gland itself (8,9). Regardless of the site of action, our results suggest that the acute administration of hydrocortisone, while effectively preventing the ascorbic acid depletion response to stress, acts merely to depress the basal level, or physiological "set" from which the corticoid component of the stress mechanism is 0perating.t This interpretation is further supported by the absolute increase in corticosterone concentration which occurred following stress in both groups.…”

“…The pituitaries from these animals were analyzed for ACTH content by a modification of the plasma ACTH radioimmunoassay. The plasma method was modified to permit pituitary content determinations as follows: Pituitaries were weighed, homogenized in glacial acetic acid in a siliconized glass tissue grinder, and then extracted according to the method of Birmingham et al (6). An aliquot of the extract was carried through the immunoassay.…”

“…Pituitary ACTH content was measured by a modification [19] of the adrenal slice bioassay of Saffran and Schally, i.e., corticosterone production was determined by using a fluorescent technique [ 16], Pituitary glands were extracted in glacial acetic acid according to the method of Birmingham et al [22]. The capacity of the pituitary gland to release ACTH was assessed in vitro by utilizing a modification [23J of the CRF bioassay procedure of Saffran and Schally.…”
