Actions of histamine, secretin, and PGE2 on cyclic AMP production by isolated canine fundic mucosal cells.

Abstract: Cyclic AMP production was studied in isolated canine fundic gastric mucosal cells. Histamine, prostaglandin E2 (PGE2), and secretin increased cyclic AMP production by unenriched mucosal cells. In separated cell fractions, histamine stimulation of cyclic AMP production correlated with the parietal cell content of the fractions. Secretin in concentrations above 1 nM stimulated cyclic AMP production, and this effect correlated with the pepsinogen content of the separated cell fractions. At concentrations above 1 … Show more

“…The most potent stimulatory effects were observed in the smaller diameter, non-parietal cell fractions. This confirms the findings of Wollin, Soll & Samloff, (1979). PGE2 weakly stimulated cyclic AMP in the parietal cellenriched fraction while Iloprost was ineffective.…”

“…These methods and the methods for determining AP uptake have been described in detail previously (17,18). The elutriator rotor, Beckman Instruments, Inc., Spinco Div., Palo Alto, Calif. (17,22) was used to prepare fractions enriched in parietal cell content (43-70%); these fractions were used for all of the subsequent studies.…”

“…Stimulation by histamine, but not by carbachol or gastrin, was found to be accompanied by increased parietal cell generation of cyclic AMP (19). Prostaglandin E2 (PGE2), which is known to stimulate cyclic AMP production by gastric mucosa (20,21), was found to increase cyclic AMP production predominantly in nonparietal cells, with at most a very small effect on parietal cells (22). These observations with isolated mucosal cells did not resolve the question of the relation, if any, between the effects of PGE2 on cyclic AMP production and inhibition of acid secretion by PGE2.…”

“…PGE2 has been shown to stimulate the rate of bone resorption induced by the parathyroid hormone (MacDonald, 1986). In rat gastric mucosa, PGE2 causes inhibition of gastric acid secretion in the parietal cell but stimulates gastric nonparietal secretion (Reeves and Stables, 1985;Major and Scholes, 1978;Soll, 1980;Wollin et al, 1979). In the amphibian cornea, both E and F prostaglandins stimulate er transport across the epithelium, and thus contribute to the maintenance of corneal transparency (Beitch et al, 1974;Bentley and McGahen, 1982).…”
