Activation of AQP4, p66Shc and endoplasmic reticulum stress is involved in inflammation by carrageenan and is suppressed by argirein, a derivative of rhein

Abstract: Activated AQP4, endoplasmic reticulum stress and p66Shc were actively implicated in the inflammation and these were suppressed by argirein, and its activity is favorable due to synergism in combination with L-arginine.

“…Indomethacin has analgesic, antipyretic, and antiinflammatory activity that is mediated by inhibition of COX-1 and COX-2 [Nalamachu and Wortmann, 2014]. Indomethacin can also inhibit the human multidrug resistance protein MRP4, thus blocking the prostaglandin efflux transporter [Reid et al, 2003], can also decrease neutrophil migration and paw edema [Albuquerque et al, 2009], induce nitric oxide production, reduce glutathione and malondialdehyde formation, reduce NF-jB mRNA expression [El-Shitany et al, 2014] and suppress aquaporin 4, endoplasmic reticulum stress and p66Shc gene expression [Cong et al, 2012]. Additionally, the potential participation of adrenaline, adrenoceptors and prednisolone in indomethacin-induced anti-inflammatory activity has been suggested [Suleyman et al, 2008].…”

“…However, the effects of rhein on ATF-6 are not all positive. Rhein can also significantly inhibit ATF-6 activation to exert its anti-inflammatory activity (Cong et al, 2012a , b ). In general, rhein can apparently upregulate ER stress, while the regulation on ATF-6 is controversial.…”

“…9 Cocrystals can also extend the market cycle of active pharmaceutical ingredient (API), endow natural products with patent and significantly reduce the cost of research and development. 10 Though forming cocrystals can greatly improve the nature of API, only one cocrystal has appeared so far on the market as medicine (glycine-theophylline cocrystal). 11 The probable reason may be that the purity of cocrystals is difficult to confirm and literature on the topic is scarce.…”
