Activation of basal forebrain-to-lateral habenula circuitry drives reflexive aversion and suppresses feeding behavior

Abstract: Environmental cues and internal states such as mood, reward, or aversion directly influence feeding behaviors beyond homeostatic necessity. The hypothalamus has been extensively investigated for its role in homeostatic feeding. However, many of the neural circuits that drive more complex, non-homeostatic feeding that integrate valence and sensory cues (such as taste and smell) remain unknown. Here, we describe a basal forebrain (BF)-to-lateral habenula (LHb) circuit that directly modulates non-homeostatic feed… Show more

“…The behavioral test was conducted in the arena under the same optogenetic stimulation paradigms as in the fMRI experiments to examine the locomotion speed, quiet wakefulness, grooming and object explorations (Figure 5A-C). The VGLUT2-Cre group exhibited a significantly higher locomotion speed (Figure 5D), which was consistent with the wakefulness and avoidance behavior-promoting effect of BF VGLUT2 neurons 3,15,16 . The ChAT-Cre group demonstrated significantly higher novel object explorations (Figure 5E), which is consistent with the involvement of cholinergic systems in attention, learning, and memory 11,12 .…”

“…Notably, the diagonal band of Broca (DBB) within the basal forebrain is one such node with roles in sensory processing, attention, motivation, and more recently, appetite regulation ( Herman et al, 2016 ; Patel et al, 2019 ). Ablation of cholinergic DBB neurons led to hyperphagia and obesity ( Herman et al, 2016 ), and activation of excitatory DBB neurons downstream of cholinergic neurons led to hypophagia and starvation ( Patel et al, 2019 ; Swanson et al, 2022 ). However, the DBB projects to numerous downstream nodes, and it is unknown which of these may drive the appetite suppression elicited by DBB activation.…”
