Activation of the Cyclin-dependent Kinase CTDK-I Requires the Heterodimerization of Two Unstable Subunits

Abstract: RNA polymerase II CTD kinases are key elements in the control of mRNA synthesis. They constitute a family of cyclin-dependent kinases activated by C-type cyclins. Unlike most cyclin-dependent kinase complexes, which are composed of a catalytic and a regulatory subunit, the yeast CTD kinase I complex contains three specific subunits: a kinase subunit (Ctk1), a cyclin subunit (Ctk2), and a third subunit (Ctk3) of unknown function that does not exhibit any similarity to known proteins. Like the Ctk2 cyclin that i… Show more

“…CTK1, a yeast CDK9 orthologue, forms a ternary complex with a cyclin, CTK2, and a third partner, CTK3 (55). In contrast to MAQ1, MAT1 and CTK3 are required for proper kinase activity of CDK7 and CTK1, respectively (10,23). Another group of CDK regulators consists of CDK inhibitors such as p21/Cip1, p27/Kip1, p57Kip2, and Ink4, which bind to and inhibit CDK2-, CDK4-, and CDK6-cyclin complexes (11,40,57).…”

“…Since P-TEFb belongs to the large Cdk-cyclin family, it is difficult to find orthologs of P-TEFb based solely on the sequence information. However, recent genetic and biochemical evidence from yeast suggests two Cdkcontaining complexes, Ctk1/Ctk2/Ctk3 and Bur1/Bur2, as possible counterparts of P-TEFb (8,21). If there is no NELF, FIG.…”

“…27 Ctk1 forms a complex with its cyclin partner, Ctk2, and Ctk3, which is important for stability of the complex. 28,29 The other budding yeast Ser2 kinase complex components, Bur1 and Bur2, were identified in a screen for factors that could bypass the need for upstream activating sequences (UAS) of a reporter gene (BUR, bypass UAS requirement 30 ). These proteins were also found to comprise a cyclin dependent kinase complex in which Bur1 is the kinase and Bur2 is the cyclin.…”
