Activation of the EGFR-PI3K-CaM pathway by PRL-1-overexpressing placenta-derived mesenchymal stem cells ameliorates liver cirrhosis via ER stress-dependent calcium

Abstract: Background Cholesterol accumulation and calcium depletion induce hepatic injury via the endoplasmic reticulum (ER) stress response. ER stress regulates the calcium imbalance between the ER and mitochondria. We previously reported that phosphatase of regenerating liver-1 (PRL-1)-overexpressing placenta-derived mesenchymal stem cells (PD-MSCsPRL−1) promoted liver regeneration via mitochondrial dynamics in a cirrhotic rat model. However, the role of PRL-1 in ER stress-dependent calcium is not clea… Show more

“…The EGFR signaling axis has been demonstrated to be important in developing liver diseases such as cirrhosis and liver cancer [ 120 ]. Inhibiting EGFR reduces liver fibrosis and HSC activation in NAFLD based on recent research, indicating the dysregulation of EGFR in animal models of liver damage [ 121 , 122 ]. EGFR is phosphorylated in the liver tissues of high-fat diet (HFD) mice with NAFLD.…”

“…Nrf2 plays an important role in maintaining the structural integrity and functional stability of mitochondria [ 115 ]. Phosphatase of regenerating liver-1 (PRL-1)-overexpressing MSC decreased endoplasmic reticulum (ER) stress via PRL-1 binding to epidermal growth factor receptor (EGFR), thereby activating PI3K expression finally regulating the release of Ca 2+ in hepatic cells [ 116 ].…”

“…Therefore, identifying diagnostic biomarkers and exploring reliable drug targets can guide clinical treatment. Liver cirrhosis is associated with serum calcium levels ( Bandi et al, 1997 ; Kim et al, 2021 ). Platelet calcium ion depth was significantly lower in cirrhotic patients than in normal controls.…”
