In this study, we synthesised the zinc oxide nanoparticles from Vernonia amygdalina and evaluated its anti-inflammatory and antinociceptive potentials against the different inflammation and pain induced mice model. The synthesised zinc oxide nanoparticles were characterised by UV, SEM, XRD and FTIR techniques. The anti-nociceptive effects of V. amygdalina were examined by different stimuli e.g. acetic acid, glutamate, capsaicin, and formalin-induced nociception in mice. The anti-inflammatory effects of synthesised zinc oxide nanoparticles were assessed by air sack assessment and the level of inflammatory cytokines were studied. The muscle tension of animals were studied through open field assessment. The present study exhibited proficient antinociceptive and anti-inflammatory actions of the synthesised Zinc oxide nanoparticles from V. amygdalina. The sormulated zinc oxide nanoparticles were appreciably reduced the acetic acid, glutamate, capsaicin, and formalin-induced nociceptive responses in mice. Further the zinc nanoparticles were exhibited the potent anti-inflammatory actions via reducing the inflammatory response and pro-inflammatory cytokines level in the mice. In conclusion, the findings of this study proved the beneficial effects of zinc oxide nanoparticles from V. amygdalina against the different pain and inflammation-induced mice. Hence, it was clear that the zinc nanoparticles from V. amygdalina could be promising antinociceptive and anti-inflammatory agent in the future.

Alkaloids are a type of natural compound possessing different pharmacological activities. Natural products, including alkaloids, which originate from plants, have emerged as potential protective agents against neurodegenerative disorders (NDDs) and chronic inflammations. A wide array of prescription drugs are used against these conditions, however, not free of limitations of potency, side effects, and intolerability. In the context of personalized medicine, further research on alkaloids to unravel novel therapeutic approaches in reducing complications is critical. In this review, a systematic survey was executed to collect the literature on alkaloids and their health complications, from which we found that majority of alkaloids exhibit anti-inflammatory action via nuclear factor-κB and cyclooxygenase-2 (COX-2), and neuroprotective interaction through acetylcholinesterase (AChE), COX, and β-site amyloid precursor protein activity. In silico ADMET and ProTox-II-related descriptors were calculated to predict the pharmacological properties of 280 alkaloids isolated from traditional medicinal plants towards drug development. Out of which, eight alkaloids such as tetrahydropalmatine, berberine, tetrandrine, aloperine, sinomenine, oxymatrine, harmine, and galantamine are found to be optimal within the categorical range when compared to nicotine. These alkaloids could be exploited as starting materials for novel drug synthesis or, to a lesser extent, manage inflammation and neurodegenerative-related complications.

Introduction: Epimedium koreanum Nakai (EKN), is a well-known Chinese herbal medicine for the treatment of osteoporosis, immunosuppression, tumours and cardiovascular diseases. Comprehensive component identification is essential for elucidation of its pharmacological mechanism and quality control. However, its complex chemical composition has caused certain difficulties in the analysis of this traditional Chinese medicine (TCM). Therefore, there is an urgent need to establish a method for rapid classification and identification of EKN chemical components.Objective: To establish a method for rapid classification and identification of the main components of flavonoids, organic acids and alkaloids in EKN. Methods:The samples were analysed by ultra-high-performance liquid chromatography quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) and data postprocessing techniques. The UPLC system used a BEH C 18 column to separate the total extract of EKN. The mobile phase consisted of 0.1% formic acid in water and acetonitrile, and the EKN extract was analysed by gradient elution at a flow rate of 0.4 mL/min. In both the positive and negative ion modes, the fragment information was obtained and compared with those of the characteristic fragmentations and neutral losses described in the literature to quickly identify the target compounds.Results: Finally, we successfully screened out 51 chemical components, including 40 flavonoids, nine organic acids, and two alkaloids. Conclusion:The established method not only comprehensively analysed the chemical compositions of EKN, solved the difficult problems of analysis and identification of the complex chemical compositions of the TCM, but also further promoted the development of the application of chemical compositions of TCM.
