Acute effects of sono-activated photocatalytic titanium dioxide nanoparticles on oral squamous cell carcinoma

Abstract: Sonodynamic therapy (SDT) is a new treatment modality using ultrasound to activate certain chemical sensitizers for cancer therapy. In this study, effects of high intensity focused ultrasound (HIFU) combined with photocatalytic titanium dioxide (TiO2) nanoparticles on human oral squamous cell line HSC-2 were investigated. Viability of HSC-2 cells after 0, 0.1, 1, or 3s of HIFU irradiation with 20, 32, 55 and 73Wcm(-2) intensities in the presence or absence of TiO2 was measured immediately after the exposures i… Show more

“…Hybrid NPS exerted the highest antiangiogenic activity [63]. Sonodynamic therapy (SDT) consisted in replacement of light in PDT with ultrasound and is emerging as a possible treatment for oral cavity SCC [64]. Moosavi Nejad et al [64] evaluated the effects of photocatalytic TiO 2 NPs combined with high intensity focused ultrasound (HIFU) on oral cavity SCC cells and subcutaneous xenograft models.…”

“…In comparison with UV or NIR radiation, US penetrates tissues much more effectively as it reaches deeper depths tumor cells. The introduction of sonosensitizers into cancer cells and their subsequent exposure to US in order to generate ROS is the underlying factor of SDT [199, 200] (Fig. 2).…”

“…Moosavi Nejda et al [200] examined in vitro the influence of the US-excited TiO 2 NPs (100 μg mL −1 ) on the viability of human oral squamous carcinoma cells (HSC-2 line). The authors proved the increased intensity of US (20, 32, 55, 73 W cm −2 ) and the prolonged sonication period (0.1, 1.0, 3.0 s) enhanced the damage ratio in the human HSC-2 cells, finally leading to their necrosis at the conditions of 73 W cm −2 and 3 s. Yamaguchi et al [183] believe that TiO 2 NP-based cancer therapies will soon become a valuable tool in fighting malignant gliomas.…”

“…68 On squamous carcinoma HSC-2, TiO 2 nanoparticles were successfully used in association with short high-intensity exposure (0.1-3 seconds at 20 to 73 W/cm 2 ). 69 Titanium dioxide can also be bound with gold as hydrophilized gold-TiO 2 nanocomposites. 70 These are efficient sensitizers that entirely suppressed tumor growth in vivo.…”
