Acute electrophysiological responses of bradykinin‐stimulated human fibroblasts.

Abstract: SUMMARY1. Acute responses to bradykinin in human dermal fibroblasts were studied at 20-24°C using both the patch-clamp technique to monitor ion currents and Fura-2 fluorescence to monitor [Ca2+]1.2. During subconfluent culture, human dermal fibroblasts can express a diversity of ion channels as described in the preceding paper.3. When GTP (1 mM) was included in the pipette solution, two additional ion channel populations were transiently augmented in response to bradykinin stimulation.4. The first is a compone… Show more

“…As is shown in the companion paper (Estacion, 1991), ion channels may be involved in the signal transduction response of human fibroblasts to a specific peptide mitogen. The possibility exists that different ion channels are involved depending on whether the signal codes for proliferation or for an action such as secretion.…”

“…Experiments performed with GTPyS included in the pipette are described in the accompanying paper (Estacion, 1991). In cells that expressed inward rectifier, however, it was noticed that the inward rectifier current seemed to consistently 'wash-out' (n = 14) over a period of minutes.…”

“…Bk-induced PI metabolism has been shown to cause an increase in the intracellular Ca2+ concentration ([Ca"fIi) in various cell preparations (Estacion, 1991;Fasolato et al, 1988;Ogura ct al., 1990). Preliminary studies on cortical astrocytes have indicated that Bk is able to mobilize [Ca2+Ii in distinct sub-populations of cells (Gimpl et al, 1992;Reiser and Hamprecht, 1985).…”

“…2C). Since many types of non-neuronal cells in the nervous system are reported to be responsive to bradykinin (2,3,6), effects of bradykinin on [Ca 2+ ] i were examined. Bradykinin applied at 1 μM caused a [Ca 2+ ] i rise in the cells not responding to the high K + stimulation (Fig.…”
