The toxicity of all atomically stable metals in the periodic table, excluding Na, Mg, K, and Ca, was measured in one-week exposures using the freshwater amphipod Hyalella azteca in both Lake Ontario, Canada, and soft water (10% Lake Ontario). Metals were added as atomic absorption standards (63 metals), and also as anion salts for 10 metals. Lethal concentrations resulting in 50% mortality (LC50s) were obtained for 48 of the metals tested; the rest were not toxic at 1,000 microg/L. The most toxic metals on a molar basis were Cd, Ag, Pb, Hg, Cr (anion), and Tl, with nominal LC50s ranging from 5 to 58 nmol/L (1 to 58 nmol/L measured). These metals were followed by U, Co, Os, Se (anion), Pt, Lu, Cu, Ce, Zn, Pr, Ni, and Yb with nominal LC50s ranging from 225 to 1,500 nmol/L (88-1,300 nmol/L measured). Most metals were similarly or slightly more toxic in soft water, but Al, Cr, Ge, Pb, and U were >17-fold more toxic in soft water; Pd was less toxic in soft water. Atomic absorption (AA) standards of As and Se in acid had similar toxicity as anions, Sb was more toxic as the AA standard, and Cr and Mn were more toxic as anions. One-week LC50s for H. azteca correlate strongly with three-week LC50s and three-week effect concentrations resulting in 50% reduction in reproduction (EC50s) in Daphnia magna.

The ability of Phanerochaete chrysosporium to reduce the oxidized forms of selenium, selenate and selenite, and their effects on the growth, substrate consumption rate, and pellet morphology of the fungus were assessed. The effect of different operational parameters (pH, glucose, and selenium concentration) on the response of P. chrysosporium to selenium oxyanions was explored as well. This fungal species showed a high sensitivity to selenium, particularly selenite, which inhibited the fungal growth and substrate consumption when supplied at 10 mg L(-1) in the growth medium, whereas selenate did not have such a strong influence on the fungus. Biological removal of selenite was achieved under semi-acidic conditions (pH 4.5) with about 40 % removal efficiency, whereas less than 10 % selenium removal was achieved for incubations with selenate. P. chrysosporium was found to be a selenium-reducing organism, capable of synthesizing elemental selenium from selenite but not from selenate. Analysis with transmission electron microscopy, electron energy loss spectroscopy, and a 3D reconstruction showed that elemental selenium was produced intracellularly as nanoparticles in the range of 30-400 nm. Furthermore, selenite influenced the pellet morphology of P. chrysosporium by reducing the size of the fungal pellets and inducing their compaction and smoothness.

ForewordThis scientific progress report is organized by the three research objectives or tasks:Task 1: To measure performance of a pilot-scale constructed wetland treatment system in terms of decreases in targeted constituents (Hg, Se and As) in FGD wastewater.Task 2: To determine how the observed performance is achieved (both reactions and rates) through sequential extractions of these elements from sediments.Task 3: To measure performance in terms of decreased bioavailability of these elements (i.e. toxicity of sediments in constructed wetlands and toxicity of outflow waters from the treatment system).Within the report, each section is divided and classified as TASK 1, 2, and 3. Discussions and conclusions are included in each section, along with an overall conclusion from data collected during the full year of funding.-3- AbstractA pilot-scale wetland treatment system was specifically designed and constructed at Clemson University to evaluate removal of mercury, selenium, and other constituents from flue gas desulfurization (FGD) wastewater. Specific objectives of this research were: (1) to measure performance of a pilot-scale constructed wetland treatment system in terms of decreases in targeted constituents (Hg, Se and As) in the FGD wastewater from inflow to outflow; (2) to determine how the observed performance is achieved (both reactions and rates); and (3) to measure performance in terms of decreased bioavailability of these elements (i.e. toxicity of sediments in constructed wetlands and toxicity of outflow waters from the treatment system).Performance of the pilot-scale constructed wetland treatment systems was assessed using two criteria: anticipated NPDES permit levels and toxicity evaluations using two sentinel toxicitytesting organisms (Ceriodaphnia dubia and Pimephales promelas). These systems performed efficiently with varied inflow simulations of FGD wastewaters removing As, Hg, and Se concentrations below NPDES permit levels and reducing the toxicity of simulated FGD wastewater after treatment with the constructed wetland treatment systems. Sequential extraction procedures indicated that these elements (As, Hg, and Se) were bound to residual phases within sediments of these systems, which should limit their bioavailability to aquatic biota.Sediments collected from constructed wetland treatment systems were tested to observe toxicity to Hyalella azteca or Chironomus tetans. Complete survival (100%) was observed for H. azteca in all cells of the constructed wetland treatment system and C. tentans had an average of 91% survival over the three treatment cells containing sediments. Survival and growth of H. azteca and C. tentans did not differ significantly between sediments from the constructed wetland treatment system and controls.
