Acyl-Lipid Metabolism

Abstract: Acyl lipids in Arabidopsis and all other plants have a myriad of diverse functions. These include providing the core diffusion barrier of the membranes that separates cells and subcellular organelles. This function alone involves more than 10 membrane lipid classes, including the phospholipids, galactolipids, and sphingolipids, and within each class the variations in acyl chain composition expand the number of structures to several hundred possible molecular species. Acyl lipids in the form of triacylglycerol … Show more

“…The enrichment of the 'suberin biosynthesis' GO term is also consistent with the enrichment in 'fatty acid biosynthesis' given that suberin and associated wax biosynthesis requires de novo fatty acid synthesis (Li-Beisson et al 2013). The induction in cork of several genes related to the synthesis, export from the plastid and elongation of fatty acids (Table S6A), according to the Aralip website (http:// aralip.plantbiology.msu.edu/pathways/pathways), reinforces the involvement of all these pathways in suberin and waxes biosynthesis.…”

Similarly, the GO terms 'glycolytic process', 'pyruvate metabolism' and 'lactate metabolic process' are also enriched in cork, probably because genes belonging to these functional categories are required to supply acetylCoA groups that are necessary for the fatty acid synthesis and elongation (Li-Beisson et al 2013). Some genes related to these categories, such as the putative orthologs of Arabidopsis sucrose synthase SUS4 and the plastidial pyruvate kinase PKp1, are clearly upregulated in the cork oak outer bark (Table S6B).…”

“…One explanation could be that additional lipases are present in photosynthetic tissues, which allow TAG turnover to continue to occur in sdp1. Arabidopsis contains many genes that could potentially have this function (Li-Beisson et al, 2013). CGI58 is one candidate, since the protein has lipase activity (Ghosh et al, 2009) and the mutant accumulates TAG in its leaves to around 0.2% of dry weight (James et al, 2010), which is higher than we detected in sdp1.…”

“…Expression levels of lipid metabolism genes were compiled according to the Arabidopsis Acyl-Lipid Metabolism database (http://aralip. plantbiology.msu.edu) (Supplemental Data Set 3) (Beisson et al, 2003;Li-Beisson et al, 2013). In cases where more than one transcript hit the same AGI protein in Arabidopsis, transcript with the highest fragments per kilobase of exon per million fragments value (and presumably highest expression level) was assigned as a representative of the AGI group (Supplemental Data Set 4).…”

“…Of the identified transcripts, 74,532 were considered as expressed (more than 50 reads per transcript across six samples) (Supplemental Data Set 7), and among the expressed transcripts, 51,230 were successfully annotated. The 1744 transcripts involved in lipid metabolism were categorized based on the lipid gene list (http:// aralip.plantbiology.msu.edu) (Beisson et al, 2003;Li-Beisson et al, 2013) (Supplemental Data Set 8). Transcripts involved in fatty acid synthesis, glycerolipid synthesis, and TAG synthesis were specifically inspected (Supplemental Data Set 9).…”
