RESUMO:A atividade antioxidante, avaliada pelo método DPPH (1,1-difenil-2-picrilidrazila), e o teor em compostos fenólicos totais do extrato bruto metanólico e frações das folhas da espécie Palicourea rigida Kunth, Rubiaceae, foram quantificadas neste trabalho. Apesar da baixa atividade apresentada pelo extrato bruto (500 ppm), a fração acetato de etila apresentou atividade moderada (192 ppm) e o maior teor de fenólicos totais dentre as frações ensaiadas. Assim, a fração acetato de etila foi submetida a procedimentos cromatográficos o que resultou no isolamento dos flavonoides quercetina 3-O-β-D-glicosídeo, quercetina 3-O-soforosídeo e isoraminetina 3-glicosídeo, cujas estruturas foram elucidadas por análise espectroscópica, incluindo RMN (1D e 2D) e comparação com os dados da literatura.Unitermos: Rubiaceae, Palicourea, flavonoides, atividade antioxidante.ABSTRACT: "Flavonoids and antioxidant activity in Palicourea rigida Kunth, Rubiaceae". The antioxidant activity, evaluated by DPPH (1,1-difenil-2-picrilidrazila) method, and the determination of the total phenolic compounds of the crude methanolic extract and fractions of the Palicourea rigida Kunth, Rubiaceae, leaves were quantified in this work. Despite weak activity exhibited by crude extract (500 ppm), the fraction ethyl acetate showed moderate activity (192 ppm), and the largest value for the phenolic compounds among all the assayed fractions. Then, the ethyl acetate fraction was submitted to the chromatography procedures which led to the isolation of the flavonoid quercetin 3-O-D-glicoside, quercetin 3-O-sophoroside and isorhamnetin 3-glicoside, which had the structures elucidated by spectroscopy analysis, including RMN (1D and 2D) and comparison with literature data.

Aurone glycosides display a variety of biological activities. However, reports about glycosyltransferases (GTs) responsible for aurones glycosylation are limited. Here, the transcriptome-wide discovery and identification of an aurone glycosyltransferase with glycosidase activity is reported. Specifically, a complementary DNA (cDNA), designated as OsUGT1, was isolated from the plant Ornithogalum saundersiae based on transcriptome mining. Conserved domain (CD)-search speculated OsUGT1 as a flavonoid GT. Phylogenetically, OsUGT1 is clustered as the same phylogenetic group with a putative 5,6-dihydroxyindoline-2-carboxylic acid (cyclo-DOPA) 5-O-glucosyltransferase, suggesting OsUGT1 may be an aurone glycosyltransferase. The purified OsUGT1 was therefore used as a biocatalyst to incubate with the representative aurone sulfuretin. In vitro enzymatic analyses showed that OsUGT1 was able to catalyze sulfuretin to form corresponding monoglycosides, suggesting OsUGT1 was indeed an aurone glycosyltransferase. OsUGT1 was observed to be a flavonoid GT, specific for flavonoid substrates. Moreover, OsUGT1 was demonstrated to display transglucosylation activity, transferring glucosyl group to sulfuretin via o-Nitrophenyl-β-d-glucopyranoside (oNP-β-Glc)-dependent fashion. In addition, OsUGT1-catalyzed hydrolysis was observed. This multifunctionality of OcUGT1 will broaden the application of OcUGT1 in glycosylation of aurones and other flavonoids.

The present study aims to investigate morphology, and anatomy of Astragalus vogelii, Bassia muricata and Morettia philaeana growing in Abu Tartur region, that lies between Kharga and Dakhla Oases in Western Desert of Egypt. Polyphenols, flavonoids, fatty acids, protein-amino acids as well as antioxidant activity of these plants were detected. The stems, leaves and roots have the general anatomical features of dicotyledons and the aerial surface of the epidermal cells covered by thick protective cuticle. Multicellular trichomes could be distinguished. The root of A. vogelii exhibited anomalous secondary thickening with rough surface periderm while those of Bassia and Morettia possess normal secondary thickening and have well-defined periderm. The highest values of polyphenols and flavonoids content were recorded in Bassia and Astragalus, respectively. Six essential and seven non-essential amino acids were identified in Astragalus shoot. GLC analysis of fatty acids revealed the presence of ten fatty acids in Bassia and twelve in Morettia shoots. The methanolic extracts of the plants showed antioxidant activity.
