Adalimumab Modulates Angiogenesis in Psoriatic Skin

Abstract: Angiogenesis plays a key role in the pathogenesis of psoriasis. New blood vessel formation occurs early during plaque lesion development in psoriatic skin, and sometimes precedes disease recurrence. TNF-α, a well established mediator of inflammation in psoriasis, up-regulates the transcription of several pro-angiogenic chemokines that are over-expressed in psoriatic skin and serum, promoting microvascular modifications in psoriatic plaque. Adalimumab is a fully human monoclonal antibody that blocks the interac… Show more

“…They may contribute to the overall pathogenetic process of psoriasis by promoting hyperplasia of epidermal keratinocytes, angiogenesis, activation and infiltration of T lymphocytes, dendritic cells, neutrophils, and other types of leucocytes in the affected skin [1,7,12,18,20,22,24,34].…”

“…Differentiated T cells produce cytokines that stimulate keratinocytes to proliferate and produce proinflammatory antimicrobial peptides and cytokines which promote angiogenesis. Cytokines produced by immune cells and keratinocytes perpetuate the inflammatory process via positive feedback loops [4,7,12,13,15,23,26,27].…”

“…Several studies evaluated the impact of therapy on global metabolic condition in psoriasis patients as well. [24][25][26][27][28][29][30][31][32] Sattar et al 33 were the first to demonstrate that anti-TNF-alpha drugs can significantly decrease some cardiovascular risks markers as Lp(a), ApoA-I, SHBG and homocysteine concentration. Also Wakke et al 34 reported on the positive effects of systemic treatments in reducing dyslipidaemia, hypertension and hyperhomocysteinaemia, oxidative stress, and endothelial cell dysfunction in psoriasis patients.…”
