Cancer chemotherapy-induced cognitive impairment (chemobrain) is a major complication that affects the prognosis of therapy. Our study evaluates the nootropic-like activity of levetiracetam (LEVE) against doxorubicin (DOX)-induced memory defects using in vivo and molecular modelling. Rats were treated with LEVE (100 and 200 mg/kg, 30 days) and chemobrain was induced by four doses of DOX (2 mg/kg, i.p.). Spatial memory parameters were evaluated using an elevated plus maze (EPM) and Y-maze. Additionally, acetylcholinesterase (AChE) and the neuroinflammatory biomarkers cyclooxygenase-2 (COX-2), prostaglandin E2 (PGE2), nuclear factor-κB (NF-κB), and tumor necrosis factor-alpha (TNF-α) were analyzed using brain homogenate. PharmMapper was used for inverse docking and AutoDock Vina was used for molecular docking. LEVE treatment significantly diminished the DOX-induced memory impairment parameters in both the EPM and Y-maze. In addition, the drug treatment significantly reduced AChE, COX-2, PGE2, NF-κB, and TNF-α levels compared to DOX-treated animals. The inverse docking procedures resulted in the identification of AChE as the potential target. Further molecular modelling studies displayed interactions with residues Gly118, Gly119, and Ser200, critical for the hydrolysis of ACh. Analysis of the results suggested that administration of LEVE improved memory-related parameters in DOX-induced animals. The ‘nootropic-like’ activity could be related to diminished AChE and neuroinflammatory mediator levels.

Epilepsy is defined as a chronic disorder that causes a sudden rush of electrical activities in the brain. These seizures may be partial or general. While fractional annexations affect a solitary part of the brain, comprehensive seizures distress the entire brain and its activities. There exist myriad options for seizure treatment. Use of Keppra, the brand name for Levetiracetam, is one of the most adopted forms of treatment. These seizures are generally caused by low blood sugar, alcohol withdrawal as well as head trauma. For one to be prescribed seizure drugs, signs such as simple partial seizures, complex partial seizures as well as general seizures must be present. There are other options for managing seizures such as brain surgery, adopting a Ketogenic diet and anti-epileptic drugs. Levetiracetam is typically prescribed as an add-on to these seizure drugs to boost their effectiveness and as well ensure improved quality of life. While essential resources and information are recommending the use of Levetiracetam, there are also sources discrediting the drug with evidence of causing memory loss and reduced quality of life. Therefore, this study aims at collecting information from different scholarly articles, journals, and websites about the side effects of using Levetiracetam in different populations. After the data is explored, the study offers a comprehensive conclusion based on the information available. The study, therefore, affirms that the use of Levetiracetam improves the quality of life and it does not affect or interfere with the cognitive functions of the brain. The study seeks to demystify the linking of Levetiracetam to memory loss of its users.

Introduction: Epilepsy and antiepileptic drugs can affect negatively the cognitive abilities of patients. Aim: The present study aimed to evaluate the effect of topiramate (TPM) and lacosamide (LCM) on the emotional and cognitive re-sponses in naive animals and in animals with pilocarpine-induced status epilepticus.  Materials and methods: Male Wistar rats were randomly divided into 6 groups and status epilepticus was evoked in half of them by a single i.p. administration of pilocarpine (Pilo) (320 mg/kg): Pilo-veh, Pilo-TPM (80 mg/kg) and Pilo-LCM (30 mg/kg). Matched naive rats were treated with the same doses as follows: C-veh, C-TPM, and C-LCM. In a step-down passive avoidance test, the learning session was held for one day, the early retention test was conducted on day 2, and the long-term memory test - on day 7. Motor activity and anxiety were evaluated in an open field test.  Results: The Pilo-TPM and Pilo-LCM groups increased the time spent on the platform compared to Pilo-veh animals while the C-LCM animals decreased the time compared to C-veh animals during short- and long-term memory retention tests. TPM and LCM exerted an anxiolytic effect in naive rats. The two antiepileptic drugs were unable to alleviate the hyperactivity, but they alleviated the impulsivity associated with decreased anxiety level in epileptic rats. Conclusions: Our findings suggest that LCM and TPM have a beneficial effect on cognition both in naive and epileptic rats. While the two antiepileptic drugs can produce an anxiolytic effect in naive rats, they alleviate the impulsivity after pilocarpine treatment.
