Adenosine A2b receptors evoke interleukin-8 secretion in human mast cells. An enprofylline-sensitive mechanism with implications for asthma.

Abstract: Adenosine potentiates mast cell activation, but the receptor type and molecular mechanisms involved have not been defined. We, therefore, investigated the effects of adenosine on the human mast cell line HMC-1. Both the A2. selective agonist CGS21680 and the A2./Anb nonselective agonist 5'-N-ethylcarboxamidoadenosine (NECA) increased cAMP, but NECA was fourfold more efficacious and had a Hill coefficient of 0.55, suggesting the presence of both A2. and Azb receptors. NECA 10 ,uM evoked IL-8 release from HMC-1,… Show more

“…The A 2B AR is positively coupled to both adenylyl cyclase and PLC [17][18][19][20] . Results indicate that the activation of PLC, through G q proteins, mediates many of the important functions of A 2B ARs 21,22 .…”

“…Activation of the A 2B AR by the non-selective agonist NECA increased inositol phosphate formation in human mast cell line HMC-1 (REF. 20), which is not sensitive to cholera or pertussis toxin but is antagonized by the slightly A 2B AR-selective antagonist enprofylline (3-propylxanthine) 20 . The arachidonic acid pathway was also recently demonstrated to be involved in A 2B AR activation 23 .…”

“…However, the A2 A family of adenosine receptors signal through cAMP and are not coupled to PLC/IP3 Schulte and Fredholm, 2003); thus, A2 B receptors would not be detected in the calcium-imaging experiments described above. The A2 B subclass of adenosine receptors are also positively coupled to cAMP, and can mediate PLC/IP3-dependent increases in intracellular calcium (Ca i 2+ ) in some cell types (Feoktistov and Biaggioni, 1995;Yakel et al, 1993). Using RT-PCR with specific primers for mouse A2 A and A2 B receptors, we detected adenosine receptor mRNA transcripts for both A2 receptor subtypes in mouse SCs in culture (Fig.…”
