Adhesive, Stretchable, and Spatiotemporal Delivery Fibrous Hydrogels Harness Endogenous Neural Stem/Progenitor Cells for Spinal Cord Injury Repair

Abstract: Aligned fibrous hydrogels capable of recruiting endogenous neural stem/progenitor cells (NSPCs) show great promise in spinal cord injury (SCI) repair. However, the hydrogels suffer from severe issues in close contact with the transected nerve stumps and harnessing the NSPC fate in the lesion microenvironment. Herein, we report aligned collagen-fibrin (Col-FB) fibrous hydrogels with stretchable property, adhesive behavior, and stromal cell-derived factor-1α (SDF1α)/paclitaxel (PTX) spatiotemporal delivery capab… Show more

“…Furthermore, the introduction of factor-1α (SDF1α) and paclitaxel (PTX) by the electrohydrodynamic jet printing method enables hydrogels with spatiotemporal delivery ability, which can recruit endogenous NSCs in situ for SCI repair (Figure 1A). 50 On one hand, the encapsulation of SDF1α facilitates the migration of NSCs. On the other hand, the encapsulation of PTX facilitates the differentiation of NSCs.…”

“…Certain special factors have been added to protein hydrogels to increase cell signalling. Using a novel approach to protein binding and assembly, Chen et al [203] added stromal cell-derived factor-1α (SDF1α)/paclitaxel (PTX) to collagen (Col-FB) fibre hydrogel (Figure 7a); Col-FB fibre hydrogel induced the migration of endogenous nerve stem/progenitor cells (NSPCs) over a longer distance (Figure 7b-d). After PTX incorporation, Col-FB fibre hydrogel can effectively promote the differentiation of NSPCs neurons (Figure 7e-g).…”
