Adipocytes as Anticancer Drug Delivery Depot

Abstract: Adipocytes were engineered for local delivery of anticancer therapeutics, including fatty acid and small-molecule drug. The lipolysis triggered by tumor cells promoted the release of these anticancer therapeutics. In addition, the immunogenic tumor phenotype induced by anticancer fatty acid combined with tumor-responsive doxorubicin prodrug enhanced cancer therapy.

“…As expected, adipocytes carrying an anti-cancer fatty acid and a doxorubicin prodrug showed effective anti-cancer ability. Moreover, these adipocytes also down-regulated the PD-L1 expression level in tumor cells, favoring the emergence of CD4+ and CD8+ T cell-mediated immune responses [ 122 ]. Similarly, nanoparticle engineered tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)-overexpressing adipose-derived stem cells could also work as drug-delivery vehicles for targeting and eradicating glioblastoma multiforme [ 123 ].…”

“…There are studies that are focusing on introducing adipocyte cell-based delivery platforms in cancer treatment and therapy [6,131]. At the same time, the adipocytes can be employed in other medical conditions such as regeneration and tissue engineering [132] or plastic surgery interventions [133,134].…”

“…[3] To address these limitations, polymer-drug conjugates that link anticancer agents to a polymer through labile bonds yield macromolecular prodrugs can achieve high loading capacity, enhanced water solubility and blood circulation levels, and site-specific accumulation within tumor tissues through an enhanced permeability and retention (EPR) effect. [4] The approach for specific prodrug activation relies on the endogenous tumor-associated stimuli, for example, enzymes, antigens, and aptamer-receptors. [4a] However, these strategies might be impeded by the tumor heterogeneity and the undesired expression of tumor-associated stimuli on healthy cells.…”
